An optimized digital RT-PCR (RT-dPCR) assay for the detection of human norovirus GI and GII RNA was compared with ISO 15216-conform quantitative real-time (RT-qPCR) assays in an interlaboratory study (ILS) among eight laboratories. A duplex GI/GII RT-dPCR assay, based on 15216-oligonucleotides, used a Bio-Rad QX200 platform by six Adapted Qiagen Qiacuity or ThermoFisher QuantStudio 3D were one laboratory each. The ILS comprised quantification absence matrix oyster tissue samples. On average,...

Seroprevalence data for pig herds suggested that there must be a relevant reservoir hepatitis E virus (HEV) in Switzerland. To know more about the viral presence ready-to-eat meat products, we screened pork liver sausages and raw from Swiss retail market of HEV. Testing was performed with detection method where extraction step optimized. As performance improved method, mean recovery rate mengovirus process control 24.4%, whereas HEV-inoculated sample matrices between 10.4 100% were achieved....

Abstract A real-time PCR method was recently published with a claim to be specific for the detection and identification of some genome-edited oilseed rape (OSR) lines commercialised in North America. The designed detect single base mutation AHAS1C gene, which confers herbicide tolerance. authors that is event-specific OSR line 5715 fulfils all requirements GMO analytical methods according EU regulations. We have thoroughly assessed relation minimum performance (MPR) established by European...

A molecular method for a rapid detection of viable Legionella pneumophila all serogroups in tap water samples was developed as an alternative to the reference (ISO). Legionellae are responsible Legionnaires' disease, severe pneumonia humans with high lethality. The is based on nutritional stimulation and increase precursor 16S rRNA indicator viability. For quantification, DNA detected by qPCR. This compared ISO using obtained from public sports facilities Switzerland. The sensitivity...

Hepatitis E caused by hepatitis viruses of the genotype 3 (HEV-3) is a major health concern in industrialized countries and due to its zoonotic character requires "One Health" approach unravel routes sources transmission. Knowing viral diversity present reservoir hosts, i.e., pigs but also wild boars, an important prerequisite for molecular epidemiology. The aim this study was gain primary information on HEV-3 subtypes along food chain Switzerland, as well within these subtypes. To end,...

In the last years, number of notified hepatitis E cases in humans has continuously increased Europe. Foodborne infection with zoonotic virus (HEV) genotype 3 is considered major cause this disease. Undercooked liver and raw sausages containing pigs wild boar are at high risk HEV. However, so far, no standardized method for detection HEV-RNA pig available. An international collaborative study on reproducibility involving 11 laboratories was performed an method, which consists steps sample...

Under current EU legislation, genetically modified organisms (GMOs) and derived food feed products must be authorized as GM food, feed, or seed appropriate detection methods made available for use in official controls. A Real-Time PCR method has recently been published by Chhalliyil et al. claiming to specific the identification of genome-edited oilseed rape (OSR) lines commercialized North America. In a previous study, we have independently assessed this three reference laboratories...

Vibrio spp. are a diverse group of ecologically important marine bacteria responsible for several foodborne outbreaks gastroenteritis around the world. Their detection and characterization moving away from conventional culture-based methods towards next generation sequencing (NGS)-based approaches. However, genomic relative in nature suffer technical biases arising library preparation sequencing. Here, we introduce quantitative NGS-based method that enables quantitation at limit...
