Immunization in pregnancy provides a promising contribution to globally reducing neonatal and under-five childhood mortality morbidity. Thorough assessment of benefits risks for the primarily healthy pregnant women their unborn babies is required. The GAIA project was formed response call World Health Organization concerted approach actively monitor safety vaccines immunization programs. aims improve quality outcome data from clinical vaccine trials with specific focus on needs requirements...
Consistent with expectations based on human in vitro microsomal experiments, administration of fluconazole (400 mg/day) for 6 days to six volunteers significantly reduced the cytochrome P450 (P450)-dependent metabolic clearance warfarin enantiomers. In particular, P4502C9 catalyzed 6- and 7-hydroxylation (S)-warfarin, pathway primarily responsible termination warfarin's anticoagulant effect, was inhibited by approximately 70%. The change (S)-warfarin pharmacokinetics caused dramatically...
Background Creating an ontology for COVID-19 surveillance should help ensure transparency and consistency. Ontologies formalize conceptualizations at either the domain or application level. Application ontologies cross domains are specified through testable use cases. Our case was extension of role Oxford Royal College General Practitioners (RCGP) Research Surveillance Centre (RSC) to monitor current pandemic become in-pandemic research platform. Objective This study aimed develop that can...
We evaluated a prototype polymerase chain reaction (PCR)-based assay for Chlamydia trachomatis developed by Roche Molecular Systems to detect endocervical infection in women. Of 587 samples obtained from women attending the Harborview Medical Center sexually transmitted diseases clinic, 58 (10%) were positive C. cell culture. Compared with culture, PCR method had sensitivity of 88% (51 58) and specificity 99.2% (525 529). The negative predictive values 92.7% 55) 98.7% 532), respectively....
To determine the accuracy of a recently developed polymerase chain reaction (PCR) urine assay to detect Chlamydia trachomatis urethral infection in men, we obtained swabs and first-catch from 365 men attending sexually transmitted diseases clinic. Thirty-three (9%) were infected with C. as defined by culture. Thirty-two 33 culture-positive also had PCR-positive assays. Of 332 patients culture-negative swabs, 325 PCR-negative urine. Compared chlamydia culture specimens, PCR samples thus...
Vaccination during pregnancy is increasingly being used as an effective approach for protecting both young infants and their mothers from serious infections. Drawing conclusions published studies in this area can be difficult because of the inability to compare vaccine trial results across different settings due heterogeneity definitions terms assess safety vaccines data collected such studies. The guidelines proposed document have been developed harmonize collection all phases clinical...
Background Since the early 1980s, Bahamas has experienced sequential epidemics of freebase/crack cocaine use, genital ulcer–inguinal adenopathy disease (GUD), and heterosexual HIV infection. Goal To prospectively define etiology GUD in patients at Princess Margaret Hospital during outbreaks crack GUD, infection Bahamas. Study Design In Nassau, 47 consecutive with underwent serologic testing for syphilis infections HIV, herpes simplex virus type 2 (HSV-2), Chlamydia trachomatis. Genital ulcer...
Bordetella bronchiseptica is a frequent cause of respiratory infections in animals but rarely causes disease humans. We describe patient with B. pneumonia and bacteremia that developed following bone marrow transplantation. infection persisted despite antimicrobial therapy may have progressed because the combined effects patient's underlying immunosuppression antagonism between doxycycline ciprofloxacin.

Immunization of pregnant women is a promising public health strategy to reduce morbidity and mortality among both the mothers their infants. Establishing safety efficacy vaccines generally uses hybrid design between conventional interventional study an observational that requires enrolling thousands participants detect unknown number uncommon events. Historically, enrollment in clinical research studies encountered many barriers based on risk aversion, lack knowledge, regulatory ambiguity....
Among 767 rectal isolates of Chlamydia trachomatis obtained from men over the period 1981–1991, 7 were found to be a rare lymphogranuloma venereum serovar, L1. These specimens taken five male patients in 1982–1983. Six seven available for DNA sequencing studies. All six these shared same sequence major outer membrane protein (MOMP) gene variable domains and had different MOMP sequences than did prototype L1, L2, L3 strains, suggesting point source outbreak. infected with serovar L-1...
As new vaccines are developed more vaccine coadministrations being offered to make delivery practical for health systems and patients. We compared the safety of coadministered with separate vaccination 20 by considering nine types adverse events following immunisation (AEFI).Real-life event data this observational cohort study were extracted from Oxford-Royal College General Practitioners Research Surveillance Centre children registered in database between 2008 2018. applied self-controlled...
The Accelerated Development of VAccine beNefit-risk Collaboration in Europe (ADVANCE) is a public-private collaboration aiming to develop and test system for rapid benefit-risk (B/R) monitoring vaccines using European healthcare databases. Event misclassification can result biased estimates. Using different algorithms identifying cases Bordetella pertussis (BorPer) infection as case, we aimed describe strategy quantify event misclassification, when manual chart review not feasible. Four...
Composite disease burden measures such as disability-adjusted life-years (DALY) have been widely used to quantify the population-level health impact of or injury, but application has limited for estimation adverse events following immunization. Our objective was assess feasibility adapting DALY approach estimating event burden. We developed a practical methodological framework, explicitly describing all steps involved: acquisition relative absolute risks and background incidence rates,...
The Accelerated Development of VAccine beNefit-risk Collaboration in Europe (ADVANCE) is a public-private collaboration aiming to develop and test system for rapid benefit-risk (B/R) monitoring vaccines, using existing healthcare databases Europe. objective this paper was assess the feasibility electronic estimate dose-specific acellular pertussis (aP) whole cell (wP) vaccine coverage. Seven four European countries (Denmark (n = 2), UK Spain 2) Italy 1)) participated study. Children were...
Both adequate coverage and adherence to paediatric immunisation schedules are required for optimal protection against vaccine preventable diseases. We studied the timeliness of routine vaccinations according NHS's schedule potential factors adherence. Immunisation data was obtained from Royal College General Practitioners (RCGP) Research Surveillance Centre (RSC). collected types, doses, dates all vaccines between 2008 2018: DTaP/IPV/Hib/HepB, DTaP/IPV/Hib, DTaP/IPV, dTaP/IPV, Td/IPV, MMR,...
