Fentanyl is primarily metabolized by CYP3A, but has also been suggested to act as a weak inhibitor of CYP3A. We investigated the influence CYP3A inhibition ketoconazole on pharmacokinetics intravenously administered fentanyl and effect activity. A prospective, open-label, randomized, monocentre, crossover study was conducted in 16 healthy volunteers. They received alone (5 microgram per kilogram) or plus (200 milligram orally B.I.D. over 2 days). Naloxone (2 × 0.2 i.v.) given simultaneously...

Therapeutic management of epilepsy remains a challenge, since optimal systemic antiseizure medication (ASM) concentrations do not always correlate with improved clinical outcome and minimal side effects. We tested the feasibility noninvasive real-time breath metabolomics as an extension traditional therapeutic drug monitoring for patient stratification by simultaneously drug-related drug-modulated metabolites. This proof-of-principle observational study involved 93 measurements 54 paediatric...

Background and Clinical Significance: We report on an infant with Snijders Blok–Campeau syndrome (psychomotor developmental delay, CNS malformations) a complex heart defect pulmonary arterial hypertension. Case Presentation: A DDX3X mutation encoding for RNA helicase was detected, which may suggest association between the development of vasculopathy. However, further validation is required. Conclusions: important role in vasculature.

A recent study investigating the pharmacokinetics of fentanyl in Sprague-Dawley rats suggested to be a substrate rat organic anion-transporting polypeptide Oatp. In human beings, most important OATP for many drugs is OATP1B1. Therefore, genetic variants OATP1B1 (SLCO1B1) might modulate and efficacy beings. Sixteen healthy male female volunteers, homozygous SLCO1B1*1a (genetic wild-type) (n = 11) or *15 (deficient haplotype carrying single-nucleotide polymorphisms rs2306283 rs4149056...
