Entry of antigen-specific T cells into human tumors is critical for immunotherapy, but the underlying mechanisms are poorly understood. Here, we combined high-dimensional spatial analyses with in vitro and vivo modeling to study immune infiltration multiple myeloma (MM) its precursor monoclonal gammopathy undetermined significance (MGUS). Clustered tumor growth was a feature MM not MGUS biopsies, this pattern reproduced humanized mouse models. biopsies exhibited intralesional as well...

Dexamethasone is used to manage cerebral oedema in patients with glioblastoma, despite significant drawbacks. Herting et al. show that dexamethasone reduces via inhibition of interleukin-1 signalling. They highlight a likely interaction between and immunotherapy, propose specific may be preferable for managing oedema.

BACKGROUNDPD-1 and PD-L1 have been studied interchangeably in the clinic as checkpoints to reinvigorate T cells diverse tumor types. Data for biologic effects of checkpoint blockade human premalignancy are limited.METHODSWe analyzed immunologic a clinical trial atezolizumab patients with asymptomatic multiple myeloma (AMM), precursor malignancy. Genomic signatures purified monocytes vivo were also compared those following PD-1 lung cancer patients. Effects on monocyte-derived DCs better...

Current management of childhood leukemia is tailored based on disease risk determined by clinical features at presentation. Whether properties the host immune response impact and outcome not known. Here, we combine mass cytometry, single cell genomics, functional studies to characterize BM environment in children with B acute lymphoblastic myelogenous T cells marrow demonstrate evidence chronic activation exhaustion/dysfunction, attrition naive TCF1+ stem-like memory accumulation terminally...

Background
Entry of neoantigen-specific T cells into tumors is critical for immunotherapy but underlying mechanisms are poorly understood. Spatial aspects immune infiltration impact outcome in solid tumors, the spatial patterns hematologic malignancies such as multiple myeloma unknown.
Methods
We combined several high dimensional analysis platforms with in-vitro/in-vivo modeling humanized mice human MM and its precursor monoclonal gammopathy undetermined significance to...
