We studied the ability of ECG to detect pathological changes in isoproterenol-induced remodeling rat heart. Myocardial hypertrophy rats was induced by repeated injections isoproterenol (5 mg/kg s.c. 7 days, Iso5, n=7). Single overdose (150 s.c., Iso150, n=7) evoked myocardial infarction followed with ventricular remodeling. The electrocardiograms were recorded anesthetized animals (thiopenthal 45 i.p.) and contractile performance analyzed isolated hearts perfused according Langendorff....

Abstract Cardiac excitation-contraction coupling relies on dyads, the intracellular calcium synapses of cardiac myocytes, where plasma membrane contacts sarcoplasmic reticulum and electrical excitation triggers release. The morphology dyads dynamics local release vary substantially. To better understand correspondence between structure functionality we estimated incidences structurally different kinetically sites tested their responsiveness to experimental myocardial injury in left...

In the present work, effect of isoproterenol on electrical properties rat heart and cytoarchitecture surviving cardiomyocytes was studied. Myocardial remodelling induced by daily administration 5 mg/kg (Iso) for 7 days. Administration resulted in a significant increase (52%) ratio left ventricular weight to body weight. ECG voltage criteria confirmed presence hypertrophy. QT interval prolongation 23% 58% found Iso rats corresponding isolated hearts, respectively. Spontaneously beating hearts...

In view of previously reported increased capacity for nitric oxide production, we suggested that l-arginine (ARG), the synthase (NOS) substrate, supplementation would improve cardiac function in isoproterenol (ISO)-induced heart failure. Male Wistar rats were treated with ISO 8 days (5 mg/kg/day, i.p.) or vehicle. ARG was given to control (ARG) and ISO-treated (ISO+ARG) water (0.4 g/kg/day). administration associated 40% mortality, ventricular hypertrophy, decreased rate, left dysfunction,...

The activation of the renin-angiotensin system (RAS) contributes to pathogenesis cardiac damage during diabetes. In present study, we investigated role pioglitazone, dapagliflozin and their combination on RAS components in streptozotocin-induced diabetic cardiomyopathy Wistar rats. Blood glucose, serum lipids, ACE (angiotensin-converting enzyme), ACE2 levels were determined. mRNA Myh6 (myosins heavy chain), Myh7, Ace, Ace2, Nppa, Nppb (natriuretic peptide A, B) Ppars (peroxisome proliferator...

The progression of diabetes mellitus leads in time to the development serious cardiovascular complications. Pycnogenol® (PYC) belongs strong antioxidants that may interfere with different pathways playing an important role diseases associated oxidative stress. Metformin (MET), commonly used antidiabetic drug, has cardio-protective effects via activation AMP kinase (AMPK). In our study, we examined PYC as add-on drug metformin therapy streptozotocin (STZ)-induced diabetic rats. Our results...

Tyrosine kinases inhibitors (TKIs) may alter glycaemia and be cardiotoxic with importance in the diabetic heart. We investigated effect of multi-TKI crizotinib after short-term administration on metabolic modulators heart rats. Experimental diabetes mellitus (DM) was induced by streptozotocin (STZ; 80 mg·kg –1 , i.p.), controls (C) received vehicle. Three days STZ, (STZ+CRI; 25 per day p.o.) or vehicle administered for 7 days. Blood glucose, C-peptide, glucagon were assessed plasma samples....

Abstract Background In spite of disrupted repolarization diabetic heart, some studies report less tendency heart to develop ventricular arrhythmias suggesting effective compensatory mechanism. We hypothesized that myocardial alterations in HCN2 and HCN4 channels occur under hyperglycaemia. Methods Diabetes was induced rats using a single injection streptozotocin (STZ; 55 mg/kg body weight, i.p.). Basal ECG measured. Expression mRNA for HCN channels, potassium microRNA 1 133a were measured...

Abstract Objectives Dapagliflozin (Dapa) could potentially be used to treat type 1 diabetes mellitus. We tested the hypothesis that it would influence blood lipid levels and visceral fat accumulation in a rodent diabetic model. Methods three groups of male Wistar rats: Controls, streptozotocin (STZ)-treated rats STZ-treated orally with Dapa (STZ+Dapa), 10 mg/kg/day for six weeks. Blood glucose serum lipids were determined. Plasma lipases (hormone-sensitive lipase, HSL lipoprotein LPL),...

Introduction: Endothelial dysfunction (ED) plays a key role in the pathogenesis of diabetic vascular complications. In monotherapy, dapagliflozin (Dapa) as well pioglitazone (Pio) prevent progression target organ damage both type 1 (T1DM) and 2 diabetes. We investigated whether simultaneous PPAR-γ activation SGLT2 cotransporter inhibition significantly alleviate ED-related pathological processes thus normalize response experimental T1DM....

Although rats a useful scientific tool, their hearts, unlike larger species, usually do not respond to torsadogenic drugs. Consequently, resistance drug‐induced arrhythmias is poorly understood. Here, we challenged with rapid delayed rectifier current (Ikr)‐inhibiting antibiotic clarithromycin (CLA), loop diuretic furosemide (FUR) or combination (CLA + FUR), examined...
