A novel cell-targeting, pH-sensitive polymeric carrier was employed in this study for delivery of the anticancer drug bortezomib (BTZ) to cancer cells. Our strategy is based on facile conjugation BTZ catechol-containing carriers that are designed be taken up selectively by cells through cell surface receptor-mediated mechanisms. The polymer used as a building block poly(ethylene glycol), which chosen its ability reduce nonspecific interactions with proteins and catechol moiety exploited bind...

On the spot: Carbohydrate microarrays, in concert with surface plasmon resonance, are valuable tools rapid identification of carbohydrate–protein interactions, like those between high-mannose oligosaccharides and cyanovirin-N (see picture). By utilizing chemical methods amenable to current high-throughput screening technologies, high-density carbohydrate microarrays can be prepared screened by using conventional widely accessible DNA microarray instrumentation.

Peptide and carbohydrate biochips can be assayed with MALDI mass spectrometry. Self-assembled monolayers (SAMs) that are modified ligands provide a platform for identifying selective protein–ligand enzyme–substrate interactions. provides rapid sensitive method detection avoids the need to label protein analytes fluorescent or radioactive tags. The picture shows spectrum of SAM presenting carbohydrate.

We describe the use of native chemical ligation (NCL) reaction to covalently cross-link soluble polymers into hydrogels. Macromonomers consisting a four-armed poly(ethylene glycol) (PEG) core end-functionalized with either thioester or N-terminal cysteine peptide were designed and synthesized. Upon mixing aqueous solutions macromonomers, rigid hydrogels formed within minutes. The gelation time was affected by choice buffer, pH, polymer concentration, temperature, composition conjugate....

This paper demonstrates that matrix-assisted laser desorption/ionization and time-of-flight mass spectrometry (MALDI-TOF MS) is very well suited for characterizing self-assembled monolayers (SAMs) of alkanethiolates on gold and, in particular, analyzing chemical reactions monolayers. This paper describes a label-free assay for measuring endogenous caspase protease activities in cell lysates. The format, termed SAMDI-MS (self-assembled monolayers matrix assisted laser desorption ionization time-of-flight mass spectrometry), is based on the enzymatic modification of peptides immobilized to monolayer substrates, followed by direct detection products with spectrometry. Monolayers presenting peptide substrates either caspase-3 or -8 were treated lysates from Jurkat cells that...
