NLRP3-associated autoinflammatory disease is a heterogenous group of monogenic conditions caused by NLRP3 gain-of-function mutations. The poor functional characterization most variants hinders diagnosis despite efficient anti-IL-1 treatments. Additionally, while controlled priming and activation signals, gain-of-functions have only been investigated in response to priming. Here, we characterize 34 vitro, evaluating their activity upon induction, priming, and/or sensitivity four inhibitors....

Flagellar propulsion of swimming Escherichia coli produces circling clockwise motions near planar solid surfaces. Counterclockwise motion was first reported air-TN medium interfaces, showing that slip at the interface is a key parameter bacterial swimming.

The orientation of the circular motion most swimming E. coli at a solid surface is shown to be (i) reversed by supplementing medium with alginate and (ii) restored rendering glass positively charged. Given slip promotion polymer suppression its anchoring on surface, observations are interpreted boundary. This points out bacterial as probe solid/fluid boundary condition.

Condensin shapes mitotic chromosomes by folding chromatin into loops, but whether it does so DNA-loop extrusion remains speculative. Although loop-extruding cohesin is stalled transcription, the impact of transcription on condensin, which enriched at highly expressed genes in many species, unclear. Using degrons Rpb1 or torpedo nuclease Dhp1XRN2 to either deplete displace RNAPII fission yeast metaphase cells, we show that not load condensin DNA. Instead, retains cis and hinders its ability...

Nucleic acid delivery requires vectorization for protection from nucleases, preventing clearance by the reticuloendothelial system, and targeting to allow cellular uptake. Nanovectors meeting above specifications should be safe patient, simple manufacture, display long-term stability. Our nanovectors were obtained via green process of polyelectrolyte complexation, carried out at 25 °C in water a low shear rate using chitosan (a polycationic biocompatible polysaccharide specific molar mass...

Abstract NLRP3-associated autoinflammatory disease (NLRP3-AID or CAPS) is an heterogenous group of monogenic autoinflammations associated with NLRP3 gain-of-function mutations. The poor functional characterization most variants a barrier to diagnosis although patients can be efficiently treated anti-IL-1 approaches. In addition, while inflammasome controlled by coordinated priming and activation signals, gain-of-functions have been only investigated in response priming. Here, we functionally...

Abstract Condensin shapes mitotic chromosomes by folding chromatin into loops but whether it does so DNA-loop extrusion remains speculative. While loop-extruding cohesin is stalled transcription, no conclusive evidence has been provided regarding the impact of transcription on condensin despite its conserved enrichment at highly expressed genes. Using degrons Rpb1 or torpedo nuclease Dhp1 XRN2 , we depleted displaced RNAP2 in fission yeast metaphase cells. We show that not load DNA instead...
