We heterologously produced a type II lasso peptide gene cluster that was mined from Streptacidiphilus jiangxiensis in E. coli . Streptacidin is the first documented to maintain its 3D structure with methionine as steric lock.
LanD flavoproteins catalyze oxidative decarboxylation of the C-terminal Cys residue a peptide to produce an enethiol. This enethiol is highly reactive and can be coupled with upstream dehydroamino acid through Michael addition form S-[2-aminovinyl](3-methyl)cysteine, unsaturated thioether known characteristic array C-terminally macrocyclized, ribosomally synthesized posttranslationally modified peptides (RiPPs). Based on two-stage bioinformatics mining posttranslational modifications (PTMs)...
Main observation and conclusion Eight new polycyclic phloroglucinol meroterpenoids guajamers A—H ( 1 — 8 ), a methylated benzoylphloroglucinol meroterpenoid guajamer I 9 ) representing skeleton, two known analogues 10 11 were isolated from the leaves of Psidium guajava . The structures molecules elucidated by detailed analysis spectroscopic data, those , 2 unambiguously confirmed single‐crystal X‐ray diffraction study. Structurally, compounds sesquiterpene monoterpene‐ based with rearranged...
Three new C-methylated phenylpropanoid glycosides (1, 2), a 8–4′-oxyneolignan (3), together with two known analogs (4, 5), were isolated from the rhizomes of Imperata cylindrical Beauv. var. major (Nees) C. E. Hubb. Their structures determined by spectroscopic and chemical methods. Compounds 1, 2, 5 (10 μM) exhibited pronounced hepatoprotective activity against N-acetyl-p-aminophenol (APAP)-induced HepG2 cell damage in vitro assays. Furthermore, their antioxidant activities...
Two new saponins, notoginsenosides Ng1 (1) and Ng2 (2), together with seven known compounds (3-9), were isolated from the leaves of Panax notoginseng. Their structures elucidated by UV, IR, HRESIMS, NMR experiments. Compounds 6 7 showed moderate cytotoxic activities against HCT-116, IC50 values 4.98 0.64 μmol/L, respectively.
Two new triterpene saponins, namely notoginsenoside Ng5 (1) and Ng6 (2) were isolated from the leaves of Panax notoginseng, along with five known ones. Their structures determined by chemical methods, NMR X-ray experiments. The absolute configuration compound 3 four sugar units was confirmed single crystal analysis. Compounds 2–4 6 inhibited PC12 cell damage induced serum deprivation, increased viability 58.7 ± 6.7% to 66.7 4.5%, 76.1 6.1%, 64.7 5.2% 67.2 5.0% at 10 μM, respectively.
Main observation and conclusion Ten new characteristic dihydroagarofuran sesquiterpenoids three known ones were acquired from the Celastraceae plant Tripterygium wilfordii . Their structure characterizations done through spectroscopic data, while compounds 10—12 further confirmed by single crystal diffractions. Several dihydroagarofurans protected PC12 cells against H 2 O ‐induced cytotoxicity as detected CCK8 assay. Compounds 5—7 13 significantly increased levels of reactive oxygen species...
