Ataxia telangiectasia-mutated (ATM) and ataxia telangiectasia Rad3-related (ATR) kinases are two key regulators of DNA-damage responses (DDR) that mainly activated in response to DNA double-strand breaks single-stranded damages, respectively. Seckel syndrome, a rare genetic disorder characterized by microcephaly markedly reduced body size, has been associated with defective ATR-dependent damage signaling. However, the only human ATR defect reported so far is hypomorphic splicing mutation...

Neurogenesis in the developing human cerebral cortex occurs at a particularly slow rate owing part to cortical neural progenitors preserving their progenitor state for relatively long time, while generating neurons. How this balance between and neurogenic is regulated, whether it contributes species-specific brain temporal patterning, poorly understood. Here, we show that characteristic potential of cells (NPCs) remain as they generate neurons prolonged amount time requires amyloid precursor...

The multistage process of cancer formation is driven by the progressive acquisition somatic mutations. Replication stress creates genomic instability in mammals. Using a well-defined multistep leukemia model Spi-1/PU.1 overexpression mouse and Spi-1/PU.1-overexpressing human leukemic cells, we investigated relationship between DNA replication progression. Here, using molecular combing flow cytometry methods, show that Spi-1 increases speed acting specifically on elongation rather than...

The synthesis of rRNA is a key determinant normal and malignant cell growth subject to epigenetic regulation. Yet, the epigenomic features rDNA arrays clustered in nucleolar organizer regions are largely unknown. We set out explore for first time how DNA methylation distributed on individual arrays. Here we combined immunofluorescence detection modifications with fluorescence hybridization single fibers, metaphase immuno-FISH methylation-sensitive restriction enzyme digestions followed by...

Progress in sample preparation for scRNA-seq is reported based on RevGel-seq, a reversible-hydrogel technology optimized samples of fresh cells. Complexes one cell paired with barcoded bead are stabilized by chemical linker and dispersed hydrogel the liquid state. Upon gelation ice complexes immobilized physically separated without requiring nanowells or droplets. Cell lysis triggered detergent diffusion, RNA molecules captured adjacent beads further processing reverse transcription cDNA...

A microvortex is generated around an infrared laser focus where intense ac electric field applied. The authors used this optoelectrostatic for stretching individual long DNAs. When λ-or T4-phage DNA molecules were introduced into the microvortex, they stretched focus. In addition, especially longer T4 molecules, it was possible to keep in form more than 30s. Using method, length of can be measured without fixing a substrate. This method applied about 10μm.

Abstract We demonstrate an effective method for DNA immobilization on a hydrophobic glass surface. The new immobilizing technique is extremely simple compared with conventional techniques that require heterobifunctional crosslinking reagent between and substrate surface are both modified chemically. In the current studies, we designed a new approach for sizing and isolating chromosomal DNA using coil-globule transition, which avoids fragmentation of giant due to mechanical stress. Although transition is reversible globular tolerant stress, cannot be manipulated by an electric field because loss its negative charges. our system, however, extracted from agarose gel in coiled state into solution PEG, induced cations generated at anode. Abstract The approximately 16 billion neurons of the human neocortex are derived from a relatively limited number developmental neural stem cells (NSCs). During embryogenesis, cortical NSCs initially generate at particularly slow rate while preserving their progenitor state for long time. How this balance between and neurogenic is regulated, whether it contributes to species-specific brain patterning, poorly understood. Here we show that characteristic potential remain in as they prolonged...

First, and Rh1 were mixed the binding reaction was checked by electrophoresis complex. The result showed that some molecules labeled Rh1. AFM observation also overlapped molecules. It suggests could be cross-linked on points.
