The evolution of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has resulted in the emergence new variant lineages that have exacerbated COVID-19 pandemic. Some those variants were designated as concern/interest (VOC/VOI) by national or international authorities based on many factors including their potential impact vaccine-mediated protection from disease. To ascertain and rank risk VOCs VOIs, we analyze ability 14 (614G, Alpha, Beta, Gamma, Delta, Epsilon, Zeta, Eta, Theta,...

Clade 2.3.4.4b highly pathogenic avian influenza (HPAI) A(H5N1) viruses that are responsible for devastating outbreaks in birds and mammals pose a potential threat to public health. Here, we evaluated their susceptibility antivirals. Of 1,015 sequences of HPAI collected the United States during 2022, eight (∼0.8%) had molecular marker drug resistance an FDA-approved antiviral: three adamantane-resistant (M2-V27A), four oseltamivir-resistant (NA-H275Y), one baloxavir-resistant (PA-I38T)....

I. Abstract After nearly a decade without reported human A/H5N1 infections, Cambodia faced sudden resurgence with 16 cases between February 2023 and August 2024, all caused by A/H5 clade 2.3.2.1c viruses. Fourteen involved novel reassortant virus gene segments from both 2.3.4.4b The emergence of this genotype underscores the persistent ongoing threat avian influenza in Southeast Asia. This study details timeline genomic epidemiology these infections related poultry outbreaks Cambodia.

The global spread of the highly pathogenic avian influenza (HPAI) A(H5N1) virus poses a serious pandemic threat, necessitating swift development effective vaccines. success messenger RNA (mRNA) vaccine technology in COVID-19 pandemic, marked by its rapid and scalability, demonstrates potential for addressing other infectious threats, such as HPAI A(H5N1). We therefore evaluated mRNA candidates targeting panzootic A(H5) clade 2.3.4.4b viruses, which have been shown to infect range mammalian...

Oas1b was previously identified as the product of Flv(r) allele that confers flavivirus-specific resistance to virus-induced disease in mice by an uncharacterized, RNase L-independent mechanism. To gain insights about mechanism which specifically reduces efficiency flavivirus replication, cellular protein interaction partners were and their involvement Oas1b-mediated analyzed. Initial difficulties getting two-hybrid assay work with full-length led discovery this Oas uniquely has a C-terminal...

During 2023-2024, highly pathogenic avian influenza A(H5N1) viruses from clade 2.3.2.1c caused human infections in Cambodia and 2.3.4.4b the Americas. We assessed susceptibility of those to approved investigational antiviral drugs. Except for 2 isolated Cambodia, all were susceptible M2 ion channel-blockers cell culture-based assays. In neuraminidase inhibition assay, displayed inhibitor drugs oseltamivir, zanamivir, peramivir, laninamivir, AV5080. Oseltamivir was ≈4-fold less potent at...

Significance All members of the order Nidovirales, including Simian hemorrhagic fever virus (SHFV), produce subgenomic mRNAs (sg mRNAs) for their 3′ genes regulated by genomic transcription regulatory sequences (TRSs). We used a next-generation sequencing–facilitated approach to comprehensively analyze nidovirus sg mRNA transcriptome. The discovery high redundancy individual and multiple previously unreported encoding nonstructural proteins, alternative reading frame or C-terminal peptides...

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has evolved into numerous lineages with unique spike mutations and caused multiple epidemics domestically globally. Although COVID-19 vaccines are available, new variants the capacity for immune evasion continue to emerge. To understand characterize evolution of circulating SARS-CoV-2 in U.S., Centers Disease Control Prevention (CDC) initiated National Strain Surveillance (NS3) program received thousands clinical specimens from...

Influenza A(H5N1) viruses have been detected in US dairy cow herds since 2024. We assessed the pathogenesis, transmission, and airborne release of A/Michigan/90/2024, an H5N1 isolate from a farm worker Michigan, ferret model. Results show this virus caused transmission with moderate pathogenicity, including limited extrapulmonary spread, without lethality.
