Abstract Activated murine macrophages metabolize arginine by two alternative pathways involving the enzymes inducible NO synthase (iNOS) or arginase. The balance between is competitively regulated Th1 and Th2 T helper cells via their secreted cytokines: induce iNOS, whereas Whereas role of expressing iNOS as inflammatory well established, functional competence arginase remains a matter speculation. Two isoforms mammalian arginases exist, hepatic I extrahepatic II. We investigated regulation...

Abstract Resveratrol (RES), a chemopreventive molecule, inhibits the proliferation of tumor cells different etiologies. We previously showed that RES alters cell cycle and induces apoptosis in MCF‐7 breast by interfering with estrogen receptor (ERaα)–dependent phosphoinositide 3‐kinase (PI3K) pathway. Here, we analyzed signaling downstream PI3K, to understand mechanisms RES‐induced apoptosis. Apoptotic death was mediated Bcl‐2 downregulation since overexpression this protein abolished...

Increasing the thermogenic capacity of adipose tissue to enhance organismal energy expenditure is considered a promising therapeutic strategy combat obesity. Here, we report that expression p38 MAPK activator MKK6 elevated in white obese individuals. Using knockout animals and shRNA, show Mkk6 deletion increases tissue, protecting mice against diet-induced obesity development diabetes. Deletion T3-stimulated UCP1 adipocytes, thereby increasing their capacity. Mechanistically, demonstrate...

SAPK3 (stress-activated protein kinase-3, also known as p38γ) is a member of the mitogen-activated kinase family; it phosphorylates substrates in response to cellular stress, and has been shown bind through its C-terminal sequence PDZ domain α1-syntrophin. In present study, we show that SAP90 [(synapse-associated 90; PSD-95 (postsynaptic density-95)] novel physiological substrate for both SAPK3/p38γ ERK (extracellular-signal-regulated kinase). binds preferentially third residues Thr287...

Acute treatment with valproate and Li+ was found to protect cultured cerebellar granule cells against apoptosis induced by low K+ (5 mM). Because the protection unaffected MK801 (N-methyl-D-aspartate receptor inhibitor), an increase in glutamate release cannot be responsible for observed neuroprotection. Insulin also protects low-K+-induced of cells. This is totally dependent on LY294002 (a phosphatidylinositol 3-kinase inhibitor). These results suggest a role neuroprotection insulin....

Lithium protects cerebellar granule cells from apoptosis induced by low potassium, and also other apoptotic stimuli. However, the precise mechanism which this occurs is not understood. When were switched to potassium medium, activation of caspase 3 was detected within 6 h, suggesting a role in mediating under conditions potassium. In same conditions, lithium (5 m ) inhibited As did inhibit activity vitro , these results suggest that ion inhibits an upstream component required for activation....

PRIMARY cultures of cerebellar granule neurons, maintained in a serum-containing medium, underwent apoptosis when exposed to C2-ceramide, as assessed by mitochondrial reduction MTT and intranucleosomal DNA fragmentation. After an 8h exposure 50 μM cell viability decreased 25–40%. Addition lithium together with C2-ceramide resulted partial protection apoptosis, which was maximal ab mM (37% protection). When added h before the apoptotic stimulus neuroprotective effect ion clearly increased...

Here we report that, response hyperosmotic stress, also regulates formation complexes between hDlg and nuclear polypyrimidine tract-binding protein-associated-splicing factor (PSF).
