Observational studies of the ongoing coronavirus disease 2019 (COVID-19) outbreak suggest that a 'cytokine storm' is involved in pathogenesis severe illness. However, molecular mechanisms underlying altered pathological inflammation COVID-19 are largely unknown. We report here toll-like receptor (TLR) 4-mediated inflammatory signaling molecules upregulated peripheral blood mononuclear cells (PBMCs) from patients, compared with healthy controls (HC).

Nontuberculous mycobacterial pulmonary diseases (NTM-PDs) are emerging as global health threats with issues of antibiotic resistance. Accumulating evidence suggests that the gut–lung axis may provide novel candidates for host-directed therapeutics against various infectious diseases. However, little is known about in context host protective immunity to identify new NTM-PDs. This study was performed gut microbes and metabolites capable conferring Using metabolomics analysis sera from NTM-PD...

The orphan nuclear receptor ESRRA (estrogen related alpha) is critical in mitochondrial biogenesis and macroautophagy/autophagy function; however, the roles of intestinal function remain uncharacterized. Herein we identified that acts as a key regulator homeostasis by amelioration colonic inflammation through activation autophagic flux control host gut microbiota. Esrra-deficient mice presented with increased susceptibility to dextran sodium sulfate (DSS)-induced colitis upregulation...

Low back pain is a global health problem that originated mainly from intervertebral disc degeneration (IDD). Autophagy, negatively regulated by the phosphatidylinositol 3-kinase (PI3K)/Akt/mammalian target of rapamycin (mTOR) signaling pathway, prevents metabolic and degenerative diseases removing recycling damaged cellular components. Despite growing evidence autophagy occurs in disc, regulation still poorly understood.

Itaconate, a crucial immunometabolite, plays critical role in linking immune and metabolic functions to influence host defense inflammation. Due its polar structure, the esterified cell-permeable derivatives of itaconate are being developed provide therapeutic opportunities infectious inflammatory diseases. Yet, it remains largely uncharacterized whether have potentials promoting host-directed therapeutics (HDT) against mycobacterial infections. Here, we report dimethyl (DMI) as promising...

Abstract Infection with rapidly growing nontuberculous mycobacteria is emerging as a global health issue; however, key host factors remain elusive. Here, we investigated the characteristic immune profiles of peripheral blood mononuclear cells (PBMCs) from patients infected Mycobacteroides abscessus subsp . (Mabc) and M. subsp. massiliense (Mmass). Using an integrated analysis mRNA microRNA expression profiles, found that several inflammatory cytokines/chemokines [interleukin (IL)-1β, IL-6 ,...

Abstract Background The gut microbiota plays a pivotal role in maintaining intestinal homeostasis and mitigating inflammation. orphan nuclear receptor ESRRA (estrogen-related alpha) has been identified as critical regulator of mitochondrial function autophagy, yet its modulation health remains unclear. Methods We investigated using Esrra-deficient mice dextran sodium sulfate (DSS)-induced colitis model. Colitis severity is through survival rate body weight. Gut barrier was evaluated...

Abstract Mitochondrial function and innate immunity are intimately linked; however, the mechanisms how mitochondrion-shaping proteins regulate host defense remains largely unknown. Herein we show that mitofusin-2 (MFN2), a mitochondrial fusion protein, promotes through maintenance of aerobic glycolysis xenophagy via hypoxia-inducible factor (HIF)-1α during intracellular bacterial infection. Myeloid-specific MFN2 deficiency in mice impaired antimicrobial inflammatory responses against...
