Capturing mitochondria's intricate and dynamic structure poses a daunting challenge for optical nanoscopy. Different labeling strategies have been demonstrated live-cell stimulated emission depletion (STED) microscopy of mitochondria, but orthogonal are yet to be established, image acquisition has suffered either from photodamage the organelles or rapid photobleaching. Therefore, nanoscopy mitochondria largely restricted two-dimensional (2D) single-color recordings cancer cells. Here, by...
Diabetic nephropathy (DN) is one of the main causes end-stage renal disease (ESRD). Existing treatments cannot control progression diabetic very well. In nephropathy, Many monocytes and macrophages infiltrate kidney tissue. However, role these cells in pathogenesis has not been fully elucidated. this study, we analyzed patient biopsy specimens, model animals. Meanwhile, cocultured found that damaged intrinsic (glomerular mesangial tubular epithelial cells) recruited monocytes/macrophages to...
Cyclooctatetraene-conjugated cyanine dyes represent an effective strategy to improve biocompatibility under light in live-cell fluorescence imaging and analysis of mitochondria.
Abstract Mitochondria play an essential role in the life cycle of eukaryotic cells. However, we still don’t know how their ultrastructure, like cristae inner membrane, dynamically evolves to regulate these fundamental functions, response external conditions or during interaction with other cell components. Although high-resolution fluorescent microscopy coupled recently developed innovative probes can reveal this structural organization, long-term, fast and live 3D imaging remains...
We present a thiophene-based terpyridine Zn(ii) complex (namely LC) which could specifically target live cell mitochondria via binding with mitochondrial DNA (mtDNA). Due to its high biocompatibility, red emission and photo-stability, mtDNA distribution within cristae inner matrix in living cells was successfully visualized using stimulated depletion (STED) nanoscopy unprecedented resolution.
Abstract Self‐labeling protein tags can introduce advanced molecular motifs to specific cellular proteins. Here we the third‐generation covalent TMP‐tag (TMP‐tag3) and showcase its comparison with HaloTag SNAP‐tag. TMP‐tag3 is based on a proximity‐induced Michael addition between an engineered Cys of E. coli dihydrofolate reductase (eDHFR) optimized trimethoprim (TMP)–acrylamide conjugates minimal linkers. Compared previous versions, features enhanced permeability when conjugated fluorogenic...
Rhodamines have been continuously optimized in brightness, biocompatibility, and color to fulfill the demands of modern bioimaging. However, problem phototoxicity caused by excited fluorophore under long-term illumination has largely neglected, hampering their use time-lapse imaging. Here we introduce cyclooctatetraene (COT) conjugated rhodamines that span visible spectrum exhibit significantly reduced phototoxicity. We identified a general strategy for generation Gentle Rhodamines, which...
Adipose tissue plays an important role in systemic energy metabolism, and its dysfunction can lead to severe metabolic disorders. Various cells adipose communicate with each other maintain homeostasis. Extracellular vesicles (EVs) are recognized as novel medium for remote intercellular communication by transferring various bioactive molecules from parental distant target cells. Increasing evidence suggests that the endocrine functions of even homeostasis largely affected different...
Lamin A/C, a nuclear lamina protein, is essential for maintaining architecture, organizing chromatin and preserving genomic stability. However, its role in regulating DNA replication remains unclear. This study investigates how A/C regulates proper activation of origins by orchestrating the structure recruitment proliferating cell antigen (PCNA). We demonstrate that stabilizes domains (RDs) restricting mobility, spatial organization, accessibility. Furthermore, interacts with PCNA sequesters...
Objectives: Dimeric pyruvate kinase (PK) M2 (PKM2) plays an important role in promoting the accumulation of hypoxia-inducible factor (HIF)-1α, mediating aberrant glycolysis and inducing fibrosis diabetic kidney disease (DKD). The aim this work was to dissect a novel regulatory mechanism Yin Yang 1 (YY1) on lncRNA-ARAP1-AS2/ARAP1 regulate EGFR/PKM2/HIF-1α pathway DKD. Materials methods: We used adeno-associated virus (AAV)-ARAP1 shRNA knocked down ARAP1 mice overexpressed or YY1, ARAP1-AS2...
Zinc biology, featuring intertwining signaling networks and critical importance to human health, witnesses exciting opportunities in the big data era of physiology. Here, we report a class red- far-red-emitting Zn2+ probes with Kd values ranging from 190 nM 74 μM, which are particularly suitable for real-time monitoring high concentration co-released insulin during vesicular secretory events. Compared prototypical rhodamine-based probes, new exploits morpholino auxochromes eliminates...
Neuronal nuclei (NeuN) protein is a key RNA-associated to guide the transcription of RNA and regulate mRNA splicing in neurons. However, lack effective labeling tracking method has hindered elucidation biological mechanism NeuN operation living neurons understand correlated central nervous system disorders. Here, we report molecular probe that can be inserted into neighboring hydrophobic–hydrophilic region NeuN, which upon binding becomes capable emitting light red region. The specificity...
Fluorescence labeling of chemically fixed specimens, especially immunolabeling, plays a vital role in super-resolution imaging as it offers convenient way to visualize cellular structures like mitochondria or the distribution biomolecules with high detail. Despite development various distinct probes that enable super-resolved stimulated emission depletion (STED) live cells, most these membrane-potential-dependent fluorophores cannot be retained well after chemical fixation. This lack...

To make further researches on the structure-activity relationships (SARs) of our previous synthesized neonicotinoid compounds, a new series nitenpyam analogues with flexible ester arm were synthesized. Preliminary bioassays indicated that all newly designed exhibited good insecticidal activity at 100 mg/L, while 4c and 4d afforded best in vitro activity, both them had 100% mortality 20 mg/L. The SAR studies suggested their potency was dual-controlled by length size group. In addition,...

Abstract A novel series of cis ‐nitenpyram analogues ( 2a – 2p ) were designed and prepared by introducing the 1,4‐dihydropyridine, with their ‐configuration confirmed X‐ray diffraction. Preliminary bioassays showed that most compounds exhibited good insecticidal activities at 20 mg/L against Aphis medicagini , 2d afforded best activity, both them had 100% mortality 4 mg/L. In addition, molecular docking studies also performed to model ligand‐receptor complexes, results explained...
Organic fluorophores are the keystone of advanced biological imaging. The vast chemical space has been extensively explored in seek molecules with ideal properties.
