Emerging evidence indicates that ionizing radiation (IR) and chemotherapy activate Type I interferon (IFN) signaling in tumor host cells. However, the mechanism of induction is poorly understood. We identified a novel radioprotective role for DEXH box RNA helicase LGP2 (DHX58) through its suppression IR-induced cytotoxic IFN-beta [1]. inhibits activation RIG-I-like receptor (RLR) pathway upon binding viral to cytoplasmic sensors RIG-I (DDX58) MDA5 (IFIH1) subsequent IFN via mitochondrial...

Significance An undesirable outcome of radiotherapy (ionizing radiation, IR) cancer is the emergence radioresistant cells. We report that Laboratory Genetics and Physiology 2 (LGP2), a resident RIG-I (retinoic acid inducible gene I)–like receptor protein, can induce radioresistance. IR induces interferon stimulates accumulation LGP2. In turn, LGP2 shuts off synthesis blocks its cytotoxic effects. Ectopic expression enhances resistance to IR, whereas depletion effects IR. Here we show...
