Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) enters human host cells via angiotensin-converting enzyme (ACE2) and causes disease 2019 (COVID-19). Here, through a genome-wide association study, we identify variant (rs190509934, minor allele frequency 0.2-2%) that downregulates ACE2 expression by 37% (P = 2.7 × 10-8) reduces the risk of SARS-CoV-2 infection 40% (odds ratio 0.60, P 4.5 10-13), providing genetic evidence levels influence COVID-19 risk. We also replicate...

Purpose: Primary urethral adenocarcinoma (PAC) is a rare malignancy associated with poor outcomes. There are 2 primary subtypes—clear cell (CCAC) and non–clear (nCCAC). In this study, we compare oncologic outcomes between CCAC vs nCCAC histological subtypes describe differences in treatment modalities. Materials Methods: We performed retrospective review of patients diagnosed PAC at our institution 1993 to 2024. Survival analysis was stratified by histologic subtype (CCAC nCCAC) modality....

Abstract Genomic profiling studies have demonstrated that bladder cancer can be divided into two molecular subtypes referred to as luminal and basal with distinct clinical behaviors sensitivities frontline chemotherapy. We analyzed the mRNA expressions of signature genes in tumor samples from publicly available MD Anderson Cancer Center cohorts. developed a quantitative classifier transition (BLT) score which identified 80–94% sensitivity 83–93% specificity. Sarcomatoid urothelial bladder cancer (SARC) displays a high propensity for distant metastasis and is associated with short survival. We report comprehensive genomic analysis of 28 cases SARC 84 conventional carcinoma (UC), the TCGA cohort 408 muscle-invasive cancers serving as reference. SARCs show distinct mutational landscape, enrichment TP53, RB1, PIK3CA mutations. They are related to basal molecular subtype UCs could be divided into epithelial-basal more clinically aggressive...

Methylthioadenosine phosphorylase, an essential enzyme for the adenine salvage pathway, is often deficient (MTAPdef) in tumors with 9p21 loss and hypothetically renders susceptible to synthetic lethality by antifolates targeting de novo purine synthesis. Here we report our single arm phase II trial (NCT02693717) that assesses pemetrexed MTAPdef urothelial carcinoma (UC) primary endpoint of overall response rate (ORR). Three 7 enrolled patients show (ORR 43%). Furthermore, a historic cohort...

Potentially nephrotoxic drugs play an important role in acute kidney injury (AKI) children. In the previous pharmacovigilance study, we found a statistical correlation between diazepam and AKI children, but causality needs to be further verified. Cardiovascular disease (CVD) represents a significant global health challenge, characterized by high incidence rates and substantial morbidity mortality. A newer index, the Cholesterol, High-Density Lipoprotein, Glucose (CHG) has been proposed as potential diagnostic tool for metabolic disorders but not investigated its ability to predict CVD risk. This study aims evaluate predictive efficacy of CHG index in comparison well-established Triglyceride–Glucose (TyG) index. In this cohort study,...

In electron and proton radiotherapy, applications of patient‐specific bolus or compensators during radiation treatments are often necessary to accommodate patient body surface irregularities, tissue inhomogeneity, variations in PTV depths achieve desired dose distributions. Emerging 3D printing technologies provide alternative fabrication methods for these compensators. This study investigated the potential utilizing Two technologies, fused deposition modeling (FDM) selective laser sintering...

Engagement of the B cell antigen receptor initiates two concurrent processes, signaling and internalization. While both are required for normal humoral immune responses, relationship between these processes is unknown. Herein, we demonstrate that following ligation, a small subpopulation receptors inductively phosphorylated selectively retained at surface where they can serve as scaffolds assembly molecules. In contrast, larger population non-phosphorylated rapidly endocytosed. Each undergo...

Thromboxane A 2 receptor (TPr) stimulation induces cellular hypertrophy in vascular smooth muscle cells (VSMCs); however, regulation of VSMC remains poorly understood. Here we show that TPr activates AMP-activated kinase (AMPK), which turn limits TPr-induced protein synthesis VSMCs. Exposure cultured VSMCs to either agonists, IBOP and U46619, or exogenous hydrogen peroxide (H O ) caused time- dose-dependent AMPK activation, as evidenced by increased phosphorylation both AMPK-Thr172...
