Roxadustat (FG-4592) is an oral inhibitor of hypoxia-inducible factor (HIF) prolyl hydroxylase that stimulates erythropoiesis and regulates iron metabolism. In phase 2 studies involving patients with chronic kidney disease, roxadustat increased levels endogenous erythropoietin to within or near the physiologic range, along increasing hemoglobin improving homeostasis. Additional data are needed regarding efficacy safety for treatment anemia in disease who not undergoing dialysis.

FG-4592 (roxadustat) is an oral hypoxia-inducible factor (HIF) prolyl hydroxylase inhibitor (HIF-PHI) promoting coordinated erythropoiesis through the transcription HIF. Two Phase 2 studies were conducted in China to explore safety and efficacy of (USAN name: roxadustat, CDAN ), a HIF-PHI, patients with anemia chronic kidney disease (CKD), both who dialysis-dependent (DD) not (NDD). In NDD study, 91 participants randomized low (1.1–1.75 mg/kg) or high (1.50–2.25 starting doses placebo. DD 87...

Abstract Glomerular hypertension is an important factor exacerbating glomerular diseases to end-stage renal because, ultimately, it results in sclerosis (especially hypertensive and diabetic nephropathy). The precise mechanism of caused by unclear, due partly the absence suitable vitro or vivo models capable mimicking regulating complex mechanical forces and/or organ-level disease processes. We developed a “glomerulus-on-a-chip” (GC) microfluidic device. This device reconstitutes glomerulus...

Pulmonary fibrosis (PF) can severely disrupt lung function, leading to fatal consequences. Salidroside is a principal active ingredient of Rhodiola rosea and has recently been reported protect against injures. The present study was aimed at exploring its therapeutic effects on PF. Lung fibrotic injuries were induced in SD rats by single intratracheal instillation 5 mg/kg bleomycin (BLM). Then, these administrated with 50, 100, or 200 salidroside for 28 days. BLM-triggered structure...

The differentiation of fibroblasts, which are promoted by transforming growth factor-β (TGF-β)/Smad, is involved in the process pulmonary fibrosis. Rho/Rho-associated coiled-coil-forming protein kinase (Rock) pathway may regulate fibroblast and myofibroblast expression α-smooth muscle actin (α-SMA), however, mechanism not clear. aim present study was to evaluate role Rho/Rock TGF-β/Smad TGF-β1-induced lung fibroblasts differentiation. Posttranslational modification of proteins could regulate their multiple biological functions. Transforming growth factor-β receptor I and II (ALK5 TGF-βRII), which are glycoproteins, play important roles in the renal tubular epithelial-mesenchymal transition (EMT). Abstract Introduction We previously found that mesenchymal stem cells (MSCs) injected intravenously could attenuate peritoneal adhesion by secreting tumor necrosis alpha-stimulating gene (TSG)-6, while MSCs intraperitoneally not. However, the underlying mechanism remains unclear. This study was designed to investigate means which exert their effects. Methods Rat bone marrow-derived MSCs/red fluorescent protein (RFP) were either or into Sprague-Dawley (SD) rats at different time points after...
