Mesenchymal stem cell (MSC)–derived exosome plays a central role in the cell-free therapeutics involving MSCs and contents can be customized under disease-associated microenvironments. However, optimal MSC-preconditioning to enhance its therapeutic potential is largely unknown. Here, we show that preconditioning of gingival tissue-derived (GMSCs) with tumor necrosis factor-alpha (TNF-α) ideal for treatment periodontitis. TNF-α stimulation not only increased amount secreted from GMSCs, but...
Immunoregulatory properties of mesenchymal stem cell (MSC)-derived extracellular vesicles (EVs) are promising. Gingival tissue-derived MSCs (GMSCs) have unique immunoregulatory capacity and secrete large amounts EVs. Recent findings suggest that priming with inflammatory stimuli is an effective strategy for cell-free therapy. However, the precise mechanism by which contents EVs customized has not been fully elucidated. Here, we show derived from GMSCs primed a combination two...
Twenty-three strains of Rhodococcus equi from independent clinical cases were analyzed for the presence virulence plasmid DNA. Of isolates, 19 contained an 85-kb and remaining 4 a 90-kb plasmid. All isolates expressed 15- to 17-kDa antigens virulent in mice. Restriction enzyme Southern blot analyses showed large regions DNA homology between 85- plasmids. It was concluded tentatively that there are at least two plasmids R. they have common origin.
Virulent Rhodococcus equi showing 15- to 17-kDa antigens, which is virulent in mice, was found harbor an 85-kbp plasmid, and the antigens were be associated with possession of plasmid R. (S. Takai, T. Sekizaki, Ozawa, Sugawara, Y. Watanabe, S. Tsubaki, Infect. Immun. 59:4056-4060, 1991). The expression these temperature regulated: when cells grown at a low (25 32 degrees C), they did not express them, whereas expressed them large amounts higher (34 41 C). on cell surface, as evidenced by...
Calorie restriction (CR) by 30-40% decreases morbidity of age-related diseases and prolongs the lifespan various laboratory animal species. Taurine (2-aminoethanesulfonic acid) is an important nutrient for lipid metabolism as it conjugates bile acids. Here, we investigated how taurine supplementation induces effects similar to CR beneficial effects. Sprague Dawley rats were fed a diet containing different concentrations (0, 0.5, 1.0, 3.0, 5.0%) analyze on growth, blood, hepatic parameters....
Pancreatic β-cell apoptosis may be involved in the onset and progression of type 2 diabetes mellitus, although its mechanism remains unclear. We previously demonstrated that macrophage-derived interferon (IFN) β induced X-linked inhibitor apoptosis-associated factor 1 (XAF1) expression β-cells accelerated vitro. Here, we explored effects XAF1 on function vivo.Pancreatic β-cell-selective overexpressing (Xaf1 Tg) mice were generated. Xaf1 Tg their wild-type (WT) littermates fed either a normal...
Aim. The present study evaluated the therapeutic effects of luteolin in alleviating pulpitis dental pulp- (DP-) derived microvesicles (MVs) via inhibition protein kinase R- (PKR-) mediated inflammation. Methodology. Proteomic analysis immortalized human pulp (DP-1) cell-derived MVs was performed to identify PKR-associated molecules. effect on PKR phosphorylation DP-1 cells and expression tumor necrosis factor-α (TNF-α) THP-1 macrophage-like were validated. cell proliferation compared with...
Stem cell therapy for regenerative medicine has been sincerely investigated, but not still popular although some clinical trials show hopeful results. This is suggested to be a representative candidate such as bone defect due the accident, iatrogenic resection oncological tumor, congenital disease, and severe periodontitis in oral region. Recently, Bio-3D printer "Regenova®" introduced an innovative three-dimensional culture system, equipped scaffold-free bio-assembling techniques without...
Bezoars are relatively rare foreign bodies of gastrointestinal tract and often cause ileus ulcerative lesions in the stomach subsequent bleeding perforation due to their size stiffness. Therefore, removal bezoars is essential recent development devices, endoscopic procedure, applied. However, stiffness, simple failed not a few cases, surgical has also been used. Recently, efficacy combination therapy procedure dissolution using carbonated liquid reported. To develop safe effective we...
The expression profiles of exosomal microRNAs (miRNAs) are regulated by the microenvironment, and appropriate priming with mesenchymal stem cells (MSCs) is one strategies to enhance paracrine potency MSCs. Our previous work demonstrated that exosomes from tumor necrosis factor (TNF)-α-primed human gingiva-derived MSCs (GMSCs) could be a therapeutic tool against periodontitis, TNFα-inducible miR-1260b essential for inhibition alveolar bone loss. However, precise molecular mechanism underlying...
Objective This study aimed to examine the effects of a six-month group-based low-intensity resistance exercise program on depression and cognitive function hemodialysis patients.Method We conducted quasi-cluster randomized, open-label controlled from October 2017 December 2018. Forty-two patients undergoing completed trial over six months; half participated in group (n = 21, mean 74.90 years age, SD 2.23, 66.67% female) other were stretching control 72.57 2.26, 28.57% female). Abstract Amelogenin directly binds to glucose‐regulated protein 78 (Grp78). Cell migration activity is expected increase when human periodontal ligament cells (hPDLCs) overexpressing Grp78 are treated with amelogenin. Geranylgeranylacetone (GGA) a drug that induces the expression of heat shock and routinely used treat gastric ulcers. Here, we investigated changes in properties behavior hPDLCs response treatment GGA synergistic effects amelogenin stimulation pretreated for establishment novel...
Enamel matrix derivatives (EMDs)-based periodontal tissue regenerative therapy is known to promote healing with minimal inflammatory response after surgery, i. e., it promotes wound reduced pain and swelling. It has also been reported that macrophages stimulated amelogenin, a major component of EMD, produce various anti-inflammatory cytokines growth factors. We previously found stimulation monocytes murine recombinant M180 (rM180) amelogenin suppresses histocompatibility complex class II...
Severe alcoholic hepatitis (AH) has a high mortality, and it is associated with encephalopathy, acute renal failure, sepsis, gastrointestinal bleeding, endotoxemia. The 28-d mortality remains poor (34%-40%), because no effective treatment been established. Recently, corticosteroids (CS) have considered for significantly improving the prognosis of those AH, as prevents production pro-inflammatory cytokines. However, CS are not always appropriate an initial therapeutic option, such in cases...
In this study, the fraction extracted from turmeric powder with 50% ethanol and fractionated n-hexane were administered to diet-induced NASH model rats. was prepared SD rats by feeding an originally designed choline-deficient, high-fat, high-fructose (HFF-CD) diet for 10 weeks. To HFF-CD diet, hexane after fractionation added at 100 mg/kg body weight. weeks later, blood samples liver collected following parameters: lipid weights, serum ALT, AST, TG, TBARS levels, metabolism-related gene...
Lactobacillus casei subsp. 2206 exhibited much lower levels of diacetyl reductase activity than Citr + Lactococcus lactis 3022 but two-, three-, and more eightfold-higher synthase, lactate dehydrogenase, NADH oxidase activities, respectively. A requirement for metal ions by the synthases in both species was observed. The extracts strain not produced from pyruvate when reaction synthase aerated it conducted statically.
More than 80 years ago, McCay and colleagues first reported that limiting the amount of food provided to experimental animals (i.e. calorie restriction or CR) prolongs their lifespan suppresses onset progression various age-related diseases.
