NMO-IgG is a disease-specific autoantibody for neuromyelitis optica (NMO) and its target antigen aquaporin-4 (AQP4) water channel. Recently, we established sensitive anti-AQP4 antibody assay using human AQP4-transfected cells, which appeared more than the original assay. So far, there has been no large-scale study on titre in NMO related disorders. We tested 148 sera of patients with NMO, high-risk syndrome multiple sclerosis (MS), clinically isolated suggestive MS miscellaneous diseases....
Intractable hiccup and nausea (IHN) was found in eight of 47 cases relapsing neuromyelitis optica (NMO) (17%) but none 130 multiple sclerosis (MS). IHN resolved with methylprednisolone. In six cases, MRI detected linear medullary lesions involving the pericanal region, area postrema, nucleus tractus solitarius. Like long centrally located myelitis, a lesion causing may distinguish NMO from MS.
A method is described for the elicitation of substantial amounts anti-hapten and anti-protein antibodies in ascitic fluids individual mice various strains. The utilizes repeated i.p. inoculations u.5-mg quantities antigen complete Freund's adjuvant. high volume ratio adjuvant to solution, used per inoculation, were shown be critical factors. Ascitic developed after three five subsequently tapped repeatedly over a period several weeks. All strains tested produced anti-KLH antibodies, but...
We previously reported that Otx2 is essential for photoreceptor cell fate determination; however, the functional role of in postnatal retinal development still unclear although it has been to be expressed bipolar cells and photoreceptors at stages. In this study, we first examined roles terminal differentiation by analyzing Otx2; Crx double-knockout mice. Otx2+/-; Crx-/- retinas, degeneration downregulation photoreceptor-specific genes were much more prominent than suggesting a...
<h3>Background:</h3> Neuromyelitis optica (NMO) is a neurological inflammatory disease associated with autoimmunity to aquaporin 4, predominantly localised in astrocytic foot processes. Recent studies have revealed that loss of 4 and glial fibrillar acidic protein (GFAP) prominent feature NMO lesions, suggesting impairment. <h3>Objective:</h3> To reveal useful clinical biomarker NMO. <h3>Methods:</h3> Enzyme-linked immunosorbent assays were carried out for markers GFAP S100B CSFs, obtained...
Cilia function as cell sensors in many organs, and their disorders are referred to “ciliopathies.” Although ciliary components transport machinery have been well studied, regulatory mechanisms of formation maintenance poorly understood. Here we show that male germ cell-associated kinase (Mak) regulates retinal photoreceptor length subcompartmentalization. Mak was localized both the connecting cilia outer-segment axonemes cells. In -null retina, photoreceptors exhibit elongated progressive...
Autophagy is a process of bulk protein degradation and organelle turnover, current therapeutic target in several diseases. The present study aimed to clarify the significance myocardial autophagy patients with dilated cardiomyopathy (DCM). Left ventricular endomyocardial biopsy was performed 250 consecutive DCM (54.9±13.9 years; male, 79%), initially presenting decompensated heart failure (HF). association these findings HF mortality or recurrence examined. Myofilament changes, which are...
Transmural histological changes were determined morphometrically in the left ventricular free wall of 20 pigs after ligation distal anterior descending coronary artery for 10, 20, 40, and 120 minutes. Hemodynamics recorded regional blood flow distribution was measured ischemic zone. Coronary occlusion produced transmural ischemia without producing significant systemic hemodynamic change. The microsphere technique revealed that less than 0.05 ml/min per g all layers zone, i.e., inner, middle,...
The zinc finger transcription factor Blimp1 plays fundamentally important roles in many cell lineages and the early development of several types, including B T lymphocytes germ cells. Although expression developing retinal photoreceptor cells has been reported, its function remains unclear. We identified as a downstream Otx2, which an essential role fate determination. To investigate mouse retina, we ablated retina by conditional gene targeting. In knockout (CKO) number was markedly reduced...
Abstract Neuromyelitis optica ( NMO ) is characterized by severe optic neuritis and transverse myelitis. The relationship of to multiple sclerosis MS has long been debated, but classified as a demyelinating disease. Since the discovery an ‐specific autoantibody aquaporin 4 AQP 4), dominant water channel in central nervous system densely expressed on end‐feet astrocytes, clinical, magnetic resonance imaging laboratory findings distinguish from have clarified. Furthermore, pathological studies...
Background: Recently, faricimab was approved as the new drug for neovascular age-related macular degeneration (nAMD). We lack knowledge to choose between existing and this use treatment-naïve nAMD cases. In study, we compared functional morphologic effects in loading dose patients with treated either intravitreal aflibercept (IVA) or (IVF) injection a clinical setting. Method: This retrospective study included 30 eyes of 28 who started treatment IVA June September 2022 29 were administered...
Abstract Intravitreal injection of aflibercept (IVA) has successfully treated polypoidal choroidal vasculopathy (PCV), and polyp morphology is an important indicator treatment efficacy. However, many studies have not reported the presence or absence regression outcomes, few reduction outcomes in cases with residual polyps. We retrospectively measured area on indocyanine green angiography images before after IVA loading phase investigated polyps 81 eyes PCV IVA. relationship between complete...
Abstract To establish the genetic tools for conditional gene deletion in mouse retinal progenitors, we generated a Dkk3‐Cre transgenic line using bacterial artificial chromosome (BAC) transgenesis. Cre recombination efficiency vivo was assayed by crossing this line, termed BAC‐Dkk3‐Cre, with CAG‐CAT‐Z reporter line. This BAC‐Dkk3‐Cre showed recombinase activity most progenitors. detectable from embryonic day 10.5 (E10.5) and generally restricted to retina during embryogenesis. verify that...
In the vertebrate retina, Otx2 transcription factor plays a crucial role in cell fate determination of both rod and cone photoreceptors. We previously reported that conditional knockout (CKO) mice exhibited total absence rods cones retina due to their conversion amacrine-like cells. order investigate entire transcriptome CKO we compared expression profile wild-type retinas at P1 P12 using microarray. observed 101- 1049-probe sets significantly decreased P12, respectively, whereas, 3-...
To elucidate the interaction of dynamics among modules that constitute biological systems, comprehensive datasets obtained from "omics" technologies have been used. In recent plant metabolomics approaches, reconstruction metabolic correlation networks has attempted using statistical techniques. However, results were unsatisfactory and effective data-mining techniques apply appropriate are needed.Using capillary electrophoresis mass spectrometry (CE-MS) diode-array detection (CE-DAD), we...
ABSTRACT TO subgroup strains of Theiler’s murine encephalomyelitis virus (TMEV) induce a persistent central nervous system infection and demyelinating disease in mice. This serves as an experimental model multiple sclerosis (MS) because the two diseases have similar inflammatory white matter pathologies immune appears to mediate demyelination both processes. We previously reported (H. H. Chen, W. P. Wong, L. Zhang, Ward, R. Roos, Nat. Med. 1:927–931, 1995) that use alternative initiation...
Three years ago, the nonablative wrinkle reduction laser (a 585-nm laser, Chromogenex V3; Light Technologies, Llanelli, U.K.) was developed, and there have already been several reports about its clinical effectiveness. The V3 has also reported to be effective in treating acne atopic dermatitis. These results suggest that some immunological role. In this study, we investigated changes elicited by irradiation at ultrastructural level analysis of interleukin (IL)-2 IL-4 mRNA skin homing T...
