Genetic variations in the ITGAM gene (encoding CD11b) strongly associate with risk for systemic lupus erythematosus (SLE). Here we have shown that 3 nonsynonymous variants produce defective CD11b elevated levels of type I interferon (IFN-I) lupus, suggesting a direct link between reduced activity and chronically increased inflammatory status patients. Treatment small-molecule agonist LA1 led to partial integrin activation, IFN-I responses WT but not CD11b-deficient mice, protected...

Podocyte injury is an early event in diabetic kidney disease and a hallmark of glomerulopathy. MicroRNA-146a (miR-146a) highly expressed many cell types under homeostatic conditions, plays important anti-inflammatory role myeloid cells. However, its podocytes unclear. Here, we show that miR-146a expression levels decrease the glomeruli patients with type 2 diabetes (T2D), which correlates increased albuminuria glomerular damage. are also significantly reduced albuminuric BTBR ob/ob mice,...

Podocyte injury and loss mark an early step in the pathogenesis of various glomerular diseases, making these cells excellent targets for therapeutics. However, cell-based high-throughput screening assays rational development podocyte-directed therapeutics are currently lacking. Here, we describe a novel high-content screening-based phenotypic assay that analyzes thousands podocytes per condition 96-well plates to quantitatively measure dose-dependent changes multiple cellular features. Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen that causes serious infections in immunocompromised individuals and cystic fibrosis patients. ExoS ExoT are two homologous bifunctional Type III Secretion System (T3SS) virulence factors induce apoptosis target host cells. They possess GTPase Activating Protein (GAP) domain at their N-termini, which share ~76% homology, an ADP-ribosyltransferase (ADPRT) C-termini, non-overlapping substrates. Both the GAP ADPRT domains...

Magnetic resonance histology (MRH) has become a valuable tool in evaluating drug-induced toxicity preclinical models. However, its application renal injury been limited. This study tested the hypothesis that MRH could detect image-based biomarkers of chronic disease, inflammation, or age-related degeneration kidney, laying foundation for more extensive use drug toxicity. We examined entire intact kidney spontaneous model progressive nephropathy. Kidneys from male Sprague Dawley rats were...
