Ustekinumab, a fully human monoclonal antibody against interleukin-12/23, may potentially be effective for severe atopic dermatitis (AD) treatment. To evaluate efficacy and safety of ustekinumab 45 mg 90 in patients with AD.In this randomized, placebo-controlled, phase II study, Japanese (aged 20-65 years) or very AD entered 12-week double-blind treatment period during which they received (1 : 1 1) mg, placebo subcutaneous injections at weeks 0 4, follow-up until week 24. The primary end...
Abstract Our previous study showed high frequency of allelic loss at chromosome 2q37 region in oral cancer. This location contains several candidate tumor suppressor genes such as PPP1R7, ILKAP , DTYMK and ING5 . We previously 3 members inhibitor growth ( ING ) family, ING1 ING3 ING4 gene head neck As shows homology with other including highly conserved carboxy‐terminal plant homeodomain nuclear localization signal, we first picked up examined it a possible For this aim, mutation mRNA...
In tongue squamous cell carcinoma (SCC), high levels of regulatory T-cell (Treg) infiltration in tumor nests are observed the cases with poor prognosis. The role Tregs head and neck cancers remains unclear. The aim this study was to observe distribution different stages SCC estimate effects on prognosis.Thirty-four were examined immunohistochemically for CD4, CD8, Forkhead box P3 (Foxp3). Immunoreactive cells counted cancer stroma nest regions, relationships between numbers disease-free...
Abstract Background Spindle cell carcinoma (SpCC) is a rare, high malignant variant of squamous (SCC), which shows biphasic proliferation conventional SCC component and spindle shape cells with sarcomatous appearance. Methods A case bone-like calcified materials, occurring at the mandibular molar region 71-years-old Japanese male patient was presented gross finding, histological findings MRI image. To identify characteristics immunohistochemistry were performed. Results Histologically,...
Recent studies have established that, in benign tumors, a large number of cancer stem cells are present, which great implications tumor development. However, ameloblastoma, highly aggressive, locally invasive with high recurrence rate, whether or not stem-like present remains undetermined. Therefore, this study we analyzed the protein expression three candidate cell markers ameloblastoma. Immunohistochemical staining for (CSC) (CD133, CD44 and ABCG2) proliferation marker Ki-67 was performed...
Oral squamous cell carcinoma (OSCC) is thought to arise as the result of cumulative genetic or epigenetic alterations in cancer-associated genes. We focused on Dickkopf-3 (Dkk-3) gene a candidate tumor suppressor OSCC. Dkk-3 potential suppressor, and its downregulation has been reported various types malignancies. However, our previous data demonstrated that protein was dominantly expressed OSCC tissue, expression correlated with high incidence metastasis poor prognosis. In order explain...
Background: Bone morphogenic protein (BMP) and Wnt signaling pathway molecules play important roles in cytodifferentiation cell proliferation. We attempted to localize these the granular ameloblastoma. Materials methods: Four samples of paraffin‐embedded ameloblastoma with cells were studied. Immunohistochemistry was performed detect basement membrane type heparan sulfate (HS) (JM403), surface HS (10E4), heparanase, Wnt‐5a, Wnt‐2, β‐catenin, BMP‐4. Results: In all four samples, strong...
Background: Keratocystic odontogenic tumor (KCOT), also known as keratocyst, is a benign cystic neoplasm, which may be associated with nevoid basal cell carcinoma syndrome (NBCCS) and if it does, will occur multiple lesions. KCOT locally destructive despite its bland histological features. However, the neoplastic nature of not well established. Heparanase an endo‐ d ‐glucuronidase enzyme that specifically cleaves heparan sulfate (HS) increase level in tumors promotes invasion, angiogenesis,...
Ameloblastoma is the most frequently encountered odontogenic tumor, characterized by a locally invasive behavior, frequent recurrences, and, although rare, metastatic capacity. Loss or inactivation of tumor suppressor genes (TSGs) allows cells to acquire neoplastic growth. The ING family proteins are suppressors that physically and functionally interact with p53 perform important roles in apoptosis, DNA repair, cell cycle regulation, senescence. TP53 genetic alterations were reported...
We have analyzed allelic loss of the short arm chromosome 9 in 39 head and neck cancers using 13 polymorphic markers found two deletions hot spots at 9p21 9p24. Loss heterozygosity was detected least one locus 28 cases (71.8%). P16, a well-known tumor suppressor gene, is considered to be target for deletion region. However, novel frequent chromosomal candidate BRM 9p24 region, were detected. Moreover, comparison clinicopathological variables demonstrated that associated with worse prognosis.
Head and neck squamous cell carcinoma (HNSCC) is one of the most frequently occurring types cancer worldwide. We focused on fact that aberrant function Wnt/β-catenin signaling a frequent event in malignancies. Dickkopf (Dkk)-3 major negative regulator Wnt/β‑catenin signaling, which known tumor suppressor down‑regulated various cancer. However, expression profile Dkk-3 protein HNSCC has not yet been reported. The present study was conducted to investigate 90 cases tissue samples HNSCC-derived...
Microcystic adnexal carcinoma is a rare, locally aggressive cutaneous neoplasm with high probability of persistence but low metastasis. We report case 69-year-old female patient an indurated plaque at the mental region. Histologically, tumor cells invaded subcutaneous tissue and mandibular bone. The consisted mainly squamous basaloid epithelial nests cords embedded in desmoplastic stroma. A few keratin-filled microcysts ductal structures were also observed. Perineural encroachment was noted...
Head and neck squamous cell carcinoma (HNSCC) is a frequently occurring cancer, despite improvement of its treatment methods, including chemotherapy, radiotherapy, surgery, the survival remains poor. Recent advances in molecular biology human cancer indicated various abnormalities HNSCC, activation oncogenes inactivation tumor suppressor genes (TSGs). Dickkopf (Dkk)-3 gene known as negative regulator Wnt signaling suggested to function TSG several kinds malignancies. We hypothesized that...
Aims: To study the stromal variation and role of stromal–tumour cell interaction in impaired bone formation as well enhanced resorption ameloblastoma. Methods results: Four types stroma were observed histologically; fibrous, desmoplastic, myxoid with hyalinization. Osteoblast osteoclast counted using haematoxylin eosin sections immunohistochemistry CD68. After histomorphometric analysis, only fibrous distinctly identified. Secreted frizzled‐related peptide (sFRP)‐2, transforming growth...
Sirukumab, a high-affinity human monoclonal antibody that selectively binds to interleukin-6, has demonstrated efficacy in the treatment of rheumatoid arthritis (RA) global phase 1 and 2 studies. The present study evaluated safety sirukumab, as monotherapy Japanese patients with RA refractory methotrexate or sulfasalazine. In this 3, double-blind study, 122 (age ≥ 20 years) were randomized (1:1, 61 each arm) sirukumab administered subcutaneously: 50 mg once every 4 weeks (q4w) 100 (q2w)...
Background:The tumor microenvironment and its stromal cells play an important role in cancer development metastasis.Bone marrow-derived (BMDCs), a rich source of hematopoietic mesenchymal stem cells, putatively contribute to this tumoral stroma. However their characteristics roles within the are unclear. In present study, BMDCs were traced using green fluorescent protein (GFP) bone marrow transplantation model. Methods: C57BL/6 mice irradiated rescued by from GFP-transgenic mice.Lewis lung...
