HI-Glycolipid fractions normally contain P-galactosylparaglobside.An improved separation of these two glycolipids was performed as follows. The acetylated fraction applied on Silica Gel G plate immediately after heat activation (150 OC, 1 h), and developed twice in dichloroethane/methanol/water (91:0.05)with an intermediate drying. Two bands were separated revealed by spraying with Primuline (Aldrich) observed under ultraviolet light.Bands scraped, extracted chloroform/methanol(21), deacetylated.

Gangliosides are ubiquitous membrane-associated glycosphingolipids, which involved in cell growth and differentiation. Most tumor cells synthesize shed large amounts of gangliosides into their microenvironment, many studies have unraveled immunosuppressive properties. In the present study we analyzed effects GM3 GD3 gangliosides, purified from human melanoma tumors, on differentiation monocyte-derived dendritic (MoDC). At concentrations close to those detected sera patients, both...

The lipid pattern of thirteen human melanoma tumors from various tissues were investigated. In seven the tumors, an estimate was given about proportion malignant melanocytes to total cell population, and a reverse correlation determined between cells in these their neutral content. phospholipids did not show any modification, nor cholesterol cancerous tissues. ganglioside found be similar all analyzed samples, with GM3, GM2 GD3 as major components, although no level content tumors.

Human endogenous retroviruses (HERVs), remnants of ancestral viral genomic insertions, are known to represent 8% the human genome and associated with several pathologies. In particular, envelope protein HERV-W family (HERV-W-Env) has been involved in multiple sclerosis pathogenesis. Solid-phase extraction (SPE) methods are easy, rapid, and reliable. Their growing popularity is in part due to their operational simplicity cost reduction solvents, partly because they easier automate. Sphingolipids implicated various cellular events such as growth, differentiation, apoptosis. However, separation by small SPE cartridges has attracted limited attention. Here we describe an procedure on aminopropyl that sequential elution allows the of a lipid mixture into free ceramides,...

Although most glycosphingolipids (GSLs) are thought to be located in the outer leaflet of plasma membrane, recent evidence indicates that GSLs and their precursor, ceramide, also associated with intracellular organelles and, particularly, mitochondria. GSL biosynthesis starts formation ceramide endoplasmic reticulum (ER), which is transported by controversial mechanisms Golgi apparatus, where stepwise addition monosaccharides on ceramides takes place. We now report presence GSL-biosynthetic...
