HI-Glycolipid fractions normally contain P-galactosylparaglobside.An improved separation of these two glycolipids was performed as follows. The acetylated fraction applied on Silica Gel G plate immediately after heat activation (150 OC, 1 h), and developed twice in dichloroethane/methanol/water (91:0.05)with an intermediate drying. Two bands were separated revealed by spraying with Primuline (Aldrich) observed under ultraviolet light.Bands scraped, extracted chloroform/methanol(21), deacetylated.
Recent studies by our group and others have disclosed the presence of ceramides in mitochondria, activities ceramide synthase reverse ceramidase mitochondria also been reported. Since a possible contamination with ER (endoplasmic reticulum)-related compartment MAM (mitochondria-associated membrane) could not be ruled out previous studies, we re-investigated enzymes metabolism highly purified from rat liver. In present paper, show that as well are indeed able to generate vitro through both or...
Gangliosides are ubiquitous membrane-associated glycosphingolipids, which involved in cell growth and differentiation. Most tumor cells synthesize shed large amounts of gangliosides into their microenvironment, many studies have unraveled immunosuppressive properties. In the present study we analyzed effects GM3 GD3 gangliosides, purified from human melanoma tumors, on differentiation monocyte-derived dendritic (MoDC). At concentrations close to those detected sera patients, both...
The lipid pattern of thirteen human melanoma tumors from various tissues were investigated. In seven the tumors, an estimate was given about proportion malignant melanocytes to total cell population, and a reverse correlation determined between cells in these their neutral content. phospholipids did not show any modification, nor cholesterol cancerous tissues. ganglioside found be similar all analyzed samples, with GM3, GM2 GD3 as major components, although no level content tumors.
Multiple sclerosis (MS) is a complex multifactorial disease of the central nervous system (CNS) for which animal models have mainly addressed downstream immunopathology but not potential inducers autoimmunity. In absence pathogen known to cause neuroinflammation in MS, Mycobacterial lysate commonly used form complete Freund's adjuvant induce autoimmunity myelin proteins Experimental Allergic Encephalomyelitis (EAE), an model MS. The present study demonstrates that protein from human...
Human endogenous retroviruses (HERVs), remnants of ancestral viral genomic insertions, are known to represent 8% the human genome and associated with several pathologies. In particular, envelope protein HERV-W family (HERV-W-Env) has been involved in multiple sclerosis pathogenesis. Investigations detect HERV-W-Env a few other autoimmune diseases were negative, except type-1 diabetes (T1D). patients suffering from T1D, was detected 70% sera, its corresponding RNA 57% peripheral blood...
Solid-phase extraction (SPE) methods are easy, rapid, and reliable. Their growing popularity is in part due to their operational simplicity cost reduction solvents, partly because they easier automate. Sphingolipids implicated various cellular events such as growth, differentiation, apoptosis. However, separation by small SPE cartridges has attracted limited attention. Here we describe an procedure on aminopropyl that sequential elution allows the of a lipid mixture into free ceramides,...
Although most glycosphingolipids (GSLs) are thought to be located in the outer leaflet of plasma membrane, recent evidence indicates that GSLs and their precursor, ceramide, also associated with intracellular organelles and, particularly, mitochondria. GSL biosynthesis starts formation ceramide endoplasmic reticulum (ER), which is transported by controversial mechanisms Golgi apparatus, where stepwise addition monosaccharides on ceramides takes place. We now report presence GSL-biosynthetic...

In multiple sclerosis (MS), human endogenous retrovirus W family (HERV-W) envelope protein, pHERV-W ENV, limits remyelination and induces microglia-mediated neurodegeneration. To better understand its role, we examined the soluble antigen from MS brain lesions detected by specific antibodies. Physico-chemical antigenic characteristics confirmed differences between ENV syncytin-1. monomers trimers remained associated with membranes, while hexamers self-assembled into a macrostructure...
The free ceramide content of rat liver mitochondria was found to be 1.7 nmol/mg protein and outer membranes contained a three‐fold higher concentration than inner membranes. mitochondrial in neutral glycolipids 0.6 protein. long‐chain bases ceramides were d18:1 sphingosine, d18:0 3‐ketosphinganine t21:1 phytosphingosine increasing order. Sphingoglycolipids (SGLs) are cell membrane constituents. As the ceramide structure influences biological properties of SGL, we characterized by electrospray ionization tandem mass spectrometry molecular species ceramides present in SGL mouse brain. We report here for first time presence mammalian brain sphingadienine (d18:2). Sphingenine (d18:1) is all species, contrast to eicosasphingenine (d20:1), which a constituent only gangliosides. Sphingadienine galactosylceramide and sulfatides....
Gangliosides are ubiquitous, membrane-associated, glycosphingolipids, the composition and production of which is altered in many tumour cells. They have been shown to inhibit vitro generation differentiation dendritic cells (DCs) from progenitors, but their effect on human tissue-residing DCs yet be investigated. In present study, we analysed GM3 GD3 gangliosides purified melanoma tumours phenotypic functional maturation epidermal Langerhans (LCs), first immune barrier against We showed that...
Abstract Retinoblastoma is a rare tumor of the young child with an intraocular localization that leads to certain problems diagnosis. With aim defining biochemical marker—which still lacking for this disease—the gangliosides pool fresh retinoblastoma tumors were analyzed. The ganglioside pattern was shown have GM3, GM2, GM1, GD3, GD2, GD1b and GT1b as major components. occurrence high concentration GD2 in led us investigate possibility changes level sera patients, using quantitative...
The mechanisms of local anesthetic cytotoxicity remain unclear. Inhibition voltage-gated sodium channels, cytoplasmic calcium dysregulation, membrane solubilization, and apoptosis have all been suggested (1,2). Two studies, published in an abstract form, that lidocaine can induce cultured neurons (3,4). Our objective was to compare the apoptotic properties two anesthetics, ropivacaine, on cell cultures. Methods A human lymphoma T-cell line Jurkat culture maintained exponential growth RPMI...
Abstract Several studies have shown that melanoma‐associated gangliosides are immunogenic in melanoma patients and antibodies against them a favorable prognostic effect. Our study aims at characterizing the humoral immune response disease‐free, advanced vaccinated with total ganglioside fraction extracted from pooled metastases of human melanoma, containing as major GM3 GD3, minor ones GM2 GD2. Prior to vaccination, all were made disease‐free by surgical removal skin, lymph‐node or other...
