Abstract Rare genetic variants in toll-like receptor 7 (TLR7) are known to cause lupus humans and mice. UNC93B1 is a transmembrane protein that regulates TLR7 localization into endosomes. In the present study, we identify two new (T314A, located proximally domain, V117L) cohort of east Asian patients with childhood-onset systemic erythematosus. The V117L variant was associated increased expression type I interferons NF-κB-dependent cytokines patient plasma immortalized B cells. THP-1 cells...

Community-acquired pneumonia (CAP) contributes substantially to morbidity and mortality in children under the age of 5 years. In examining bronchoalveolar lavages (BALs) with CAP, we found that interleukin-17 (IL-17) production was significantly increased severe CAP. Immune profiling showed mucosal-associated invariant T (MAIT) cells from BALs, but not blood, CAP patients actively produced IL-17 (MAIT17). Single-cell RNA-sequencing revealed MAIT17 resided a BAL-resident PLZFhiCD103+ MAIT...

BackgroundMonogenic autoinflammatory disorders are characterized by dysregulation of the innate immune system, for example gain-of-function mutations in inflammasome-forming proteins, such as NOD-like receptor family CARD-containing 4 protein (NLRC4).ObjectiveHere we investigate mechanism which a novel mutation leucine-rich repeat (LRR) domain NLRC4 (c.G1965C, p.W655C) contributes to disease. Methods: We studied 2 unrelated patients with early-onset macrophage activation syndrome harboring...

The majority the compounds demonstrated vasorelaxation action on 60 mM KCl-induced contractions rat isolated aortic rings in a concentration-dependent manner, is mediated by both endothelium-independent endothelium-dependent mechanisms. Compounds 9, 12, 13, 14, 15, 19 showed higher activities comparing with lead compound 3. In addition, these had potential protective against oxLDL-induced endothelial oxidative injury...

Abstract Innate immunity is important for host defense by eliciting rapid anti-viral responses and bridging adaptive immunity. Here, we show that endogenous lipids released from virus-infected cells activate lung γδ T to produce interleukin 17 A (IL-17A) early protection against H1N1 influenza infection. During infection, the cell pool constantly supplemented thymic output, with recent emigrants infiltrating into parenchyma airway acquire tissue-resident feature. Single-cell studies identify...

This study aims to investigate clinical risk factors and early markers of E-11 associated neonatal syndrome. Methods is a multicentre retrospective cohort 105 neonates with infection China. Patients (the severe group) were compared those mild disease. Clinical analysed. In addition, cytokine analysis performed selective patients explore the immune responses. Abstract Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of disease 2019 (COVID-19), has had a significant impact on healthcare systems and economies worldwide. The continuous emergence new viral strains presents major challenge in development effective antiviral agents. Strategies that possess broad-spectrum activities are desirable to control SARS-CoV-2 infection. ACE2, an angiotensin-containing enzyme prevents overactivation renin angiotensin system, is...

Inorganic polyphosphate (poly-P), guanosine pentaphosphate (pppGpp) and tetraphosphate (ppGpp) are ubiquitous in bacteria. These molecules play a variety of important physiological roles associated with stress resistance, persistence, virulence. In the bacterial pathogen Mycobacterium tuberculosis, identities proteins responsible for metabolism (p)ppGpp remain to be fully established. M. tuberculosis encodes two PPX-GppA homologues, Rv0496 (MTB-PPX1) Rv1026, which share significant sequence...

The pathogenesis of Mycoplasma pneumoniae pneumonia (MPP), specifically the local immune responses in lungs, is poorly understood. In this study, flow cytometry was used to analyze IL-17 and related cytokines plasma bronchoalveolar lavage fluid (BALF) samples from 18 30 pediatric patients with general MPP (GMPP) refractory (RMPP), respectively. levels IL-1Ra, IL-6, IL-8, IL-17, TNF-α were significantly elevated BALF children compared (P < 0.01). Background Human adenoviruses (HAdV) infection caused pneumonia remains a major threat to global children health. Currently, diagnosis of severe HAdV in is hampered by the lack specific biomarkers. Also, severity adenovirus pediatric patients generally based on clinical features and existing biomarkers do not reliably correlate severity. Here, we asked whether local systemic inflammatory mediators could act as predicting children. Methods Totally 37 common protein levels were determined...
