To evaluate the postoperative analgesic efficacy of GW842166, a noncannabinoid CB2 agonist, in patients undergoing third molar tooth extraction. This randomized, double-blind, placebo-controlled study compared single doses GW842166 (100 or 800 mg) ibuprofen with placebo extraction at least 1 fully partially impacted tooth. Eligible participants were dosed preoperatively within hour surgery. Participants allocated to active comparator received second dose (400 mg), 4 hours after first mg dose....

The neurite outgrowth inhibitor, Nogo-A, has been shown to be overexpressed in skeletal muscle amyotrophic lateral sclerosis (ALS); it is both a potential biomarker and therapeutic target. We performed double-blind, two-part, dose-escalation study, subjects with ALS, assessing safety, pharmacokinetics (PK) functional effects of ozanezumab, humanized monoclonal antibody against Nogo-A. In Part 1, 40 were randomized (3∶1) receive single dose intravenous ozanezumab (0.01, 0.1, 5, or 15 mg/kg)...

Autotaxin-lysophosphatidic acid (ATX-LPA) signaling has a predominant role in immunological and fibrotic processes, including cancer. Several ATX inhibitors LPA receptor antagonists have been clinically evaluated, but none patients with solid tumors. Many cancers are burdened high degree of fibrosis an immune desert phenotype (so-called 'cold' tumors). In these cold tumors, the stroma provides intrinsic cancer-supporting mechanism. Furthermore, prevents penetration limits effectiveness...

Combination analgesics are effective in acute pain, and a theoretical framework predicts efficacy for combinations. The combination of dexketoprofen tramadol is untested, but predicted to be highly effective. This was randomised, double-blind, double-dummy, parallel-group, placebo-controlled, single-dose trial patients with moderate or severe pain following third molar extraction. There were ten treatment arms, including trometamol (12.5 mg 25 mg) hydrochloride (37.5 75 mg), given as four...

Background: Prognosis of patients affected by metastatic esophageal–gastric junction (EGJ) or gastric cancer (GC) remains dismal. Trastuzumab, an anti-HER2 monoclonal antibody, is the only targeted agent approved for first-line treatment with HER2-overexpressing advanced EGJ GC in combination chemotherapy. However, invariably become resistant during this treatment. We recently identified overexpression fibroblast growth factor (FGF) receptor 3 (FGFR3) as a molecular mechanism responsible...

Aims To establish whether enantioselective metabolism of racemic ( rac )‐salbutamol occurs in the lungs by determining its enantiomeric disposition following inhalation, absence and presence oral charcoal, compared with that intravenous routes. Methods Fifteen healthy subjects (eight male) were randomized into an open design, crossover study. Plasma urine salbutamol enantiomer concentrations measured for 24 h (2 mg) or without charcoal (to block absorption), inhaled (MDI; 1200 μg) (500...

ABSTRACT In a single-center, randomized study, zanamivir (Relenza) concentrations in induced sputum samples and nasal washings of healthy adults following oral inhalation were measured. Concentrations exceeded the median viral neuraminidase 50% inhibitory concentration at 6, 12, 24 h, those did so 6 12 h. There no zanamivir-related adverse events or laboratory abnormalities.
