Insulin-like growth factor-binding protein (IGFBP)-3 regulates apoptosis in an IGF-independent fashion and has been shown to localize nuclei. We cloned the nuclear receptor retinoid X receptor-alpha(RXR-alpha) as IGFBP-3 partner a yeast two-hybrid screen. Multiple methodologies showed that RXR-alpha bind each other within nucleus. IGFBP-3-induced was abolished RXR-alpha-knockout cells. RXR ligands were additive inducing prostate cancer enhanced response element inhibited RARE signaling....

Cellular stresses inhibit retinoid signaling, but the molecular basis for this phenomenon has not been revealed. Here, we present evidence that X receptor (RXR) is a substrate both mitogen-activated protein kinase kinase-4 (MKK4/SEK1) and its downstream mediator c-Jun N-terminal (JNK). MKK4/SEK1 JNK recognized distinct features on RXR in DE AB regions, respectively. Phosphorylation by had profound effects biochemical properties of RXR, inhibiting expression genes activated RXR-retinoic acid...

We have generated F9 murine embryonal carcinoma cells in which either the retinoid X receptor (RXR)α and retinoic acid (RAR)α genes or RXRα RARγ are knocked out, compared their phenotypes with those of wild-type (WT), RXRα−/−, RARα−/−, RARγ−/− cells. RXRα−/−/ RARα−/− were resistant to treatment for induction primitive parietal endodermal differentiation, as well antiproliferative apoptotic responses, whereas they could differentiate into visceral endodermlike cells, previously observed...

Glycogen synthase kinase-3β (GSK-3β) activity is suppressed when it becomes phosphorylated on serine 9 by protein kinase B (Akt). To determine how GSK-3β opposes Akt function we used various methods to alleviate suppression in prostate carcinoma cells. In some experiments, LY294002, a specific inhibitor of phosphatidylinositol 3-kinase (a involved activating Akt) and tumor necrosis factor-α (TNF-α) were activate GSK-3β. other experiments mutant forms GSK-3β, GSK-3βΔ9 constitutively active...

Retinoids inhibit the growth and enhance differentiation of murine S91-C2 melanoma cells. Specific alterations in gene expression are a plausible mechanism for these effects. Life science research is moving quickly towards large-scale experimental designs that are comprised of multiple tissues, time points, and samples.
