During development, neural circuit formation requires the stabilization of active γ-aminobutyric acid–mediated (GABAergic) synapses and elimination inactive ones. Here, we demonstrate that, although activation postsynaptic GABA type A receptors (GABAARs) adenosine A2A (A2ARs) stabilizes GABAergic synapses, only A2AR is sufficient. Both GABAAR- A2AR-dependent signaling pathways act synergistically to produce 3′,5′-monophosphate through recruitment calcium–calmodulin–adenylyl cyclase pathway....

The molecular mechanisms underlying circuit re-wiring in the mature brain remains ill-defined. An eloquent example of adult remodelling is hippocampal mossy fiber (MF) sprouting found diseases such as temporal lobe epilepsy. determinants this retrograde remain unclear. This may involve signaling system(s) controlling axon specification/growth during neurodevelopment reactivated epileptogenesis. Since adenosine A2A receptors (A2AR) control formation/outgrowth and synapse stabilization...

Cortical interneurons born in the subpallium reach cortex through tangential migration, whereas pyramidal cells their final position by radial migration. Purinergic signaling via P2Y1 receptors controls migration of intermediate precursor from ventricular zone to subventricular zone. It was also reported that blockade A2A (A2AR) somatostatin+ interneurons. Here we found A2AR control cortical projection neurons. In A2AR-knockout (KO) mouse embryos or naïve exposed an antagonist, observed...

Theobromine is a caffeine metabolite most abundant in dark chocolate, of which consumption linked with lower risk cognitive decline. However, the mechanisms through theobromine affects neuronal function remain ill-defined. Using electrophysiological recordings mouse hippocampal synapses, we now characterized impact realistic concentration on synaptic transmission and plasticity. (30 μM) facilitated while decreasing magnitude long-term potentiation (LTP), both effects being blunted by...

The human ejaculate contains subpopulations of sperm with distinct properties. Human X- and Y-bearing were separated fluorescence activated cell sorting. To avoid the use UV light quantitative DNA dyes DRAQ5® Dyecycle™ Vybrant® Violet used. Sorting efficiency was similar for both dyes, but lower than what is usually obtained classical method involving Hoechst 33342 (60-70% enrichment, versus 80-90%). A total 2,739 spermatozoa evaluated, from seven samples using in situ hybridization (FISH)...
