CD19 chimeric antigen receptor T cell (CD19CAR-T) has shown great potential to treat acute B lymphoblastic leukemia (B-ALL) and lymphoma, most of anti-CD19 scFv are derived from murine antibody sequences. However, about 10-20% B-ALL patients exhibit primary resistance murine-based CD19CAR-T (CD19mCAR-T). Herein, we report that a humanized selective (CD19hsCAR-T) may offer solution this problem.
不知火姫菊乾燥花弁を生理リン酸緩衝液(PBS)もしくはエタノールで抽出し,各種癌細胞株の増殖に及ぼす影響について検討した.いずれの抽出液も330nmに吸収極大を与え,PBS抽出液がより高い吸光度を与えた.ラット肝癌細胞株dRLh-84の培養液に10分の1量のPBS抽出液もしくは1000分の1量のエタノール抽出液を添加して4日間培養した結果,細胞増殖抑制効果はPBS抽出液にのみ認められた.PBS抽出液はラット正常肝細胞株RLN-10に対してはdRLh-84細胞より弱い増殖抑制効果を示した.各種ヒト癌細胞株に対するPBS抽出液の増殖抑制効果を比較した結果,本抽出液は肝癌細胞株HuH-7およびHepG2に対して強い毒性を示し,乳癌細胞株MCF-7にも致死作用を示したが,白血病細胞株K562およびHL-60に対しては致死効果がほとんど認められず,肺癌細胞株に対しては増殖抑制効果のみ示すことが明らかとなった.これらの結果は乾燥菊花弁抽出物の細胞毒性が癌細胞の由来により異なることを示している.
