Abstract Human infections with avian influenza H7N9 or H10N8 viruses have been reported in China, raising concerns that they might cause human epidemics and pandemics. However, how these adapt to mammalian hosts is unclear. Here we show besides the commonly recognized viral polymerase subunit PB2 residue 627 K, other residues including 87E, 292 V, 340 588 648 V 676 M also play critical roles adaptation of virus. The avian-origin H10N8, H9N2 harboring PB2-588 exhibited higher activity, more...

Senecavirus A (SVA)-induced porcine idiopathic vesicular disease has caused huge economic losses worldwide. Glucose metabolism in the host cell is essential for SVA proliferation; however, impact of virus on glucose cells and subsequent effects are still unknown. Here, glycolysis induced by shown to facilitate replication promoting lactate production, which then attenuates interaction between mitochondrial antiviral-signaling protein (MAVS) retinoic acid-inducible gene I (RIG-I). induces...

Atypical porcine pestivirus (APPV) have been detected in swine herds from the USA, Germany, Netherlands, Spain and most recently Austria, suggesting a wide geographic distribution of this novel virus. Here, for first time, we reported APPV infection China. Newborn piglets two separate Guangdong province were found showing typical congenital tremors July August 2016. RT-PCR, sequencing phylogenetic analysis showed occurred. Phylogenetic that Chinese strains, GD1 GD2, formed independent branch...

Avian influenza viruses (AIVs) are potential pandemic of global concern, posing a major threat to both the poultry industry and human health. Host factors play key role in replication AIVs, while function complement component 4 binding protein, membrane (C4BPM) this process is still unclear. This research reports that C4BPM promotes H5N1 H9N2 AIVs by inhibiting type I interferons (IFNs). infection up-regulate expression chicken embryo fibroblast cells (DF-1). Orf is a zoonotic and epitheliotrophic contagious disease that mainly affects sheep, goats, wild ruminants, humans with worldwide distribution. To date, there little information on the characterization of ORFV strains are endemic in Mainland China. In addition, relationship between severity molecular profile has not been fully elucidated. From recent outbreak sheep herd Nongan, northeast China, novel orf virus (ORFV) strain NA1/11 was successfully isolated. Western blot analysis indicated...

H9N2 subtype avian influenza viruses (AIVs) have shown expanded host range and can infect mammals, such as humans swine. To date the mechanisms of mammalian adaptation interspecies transmission AIVs remain poorly understood. explore molecular basis determining AIVs, we compared two field isolates in a mouse model: one (A/chicken/Guangdong/TS/2004, TS) is nonpathogenic, another (A/chicken/Guangdong/V/2008, V) lethal with efficient replication brains. In order to determine differences...

Although Newcastle disease virus (NDV) with high pathogenicity has frequently been isolated in poultry China since 1948, the mode of its transmission among avian species remains largely unknown. Given that various wild bird have implicated as sources transmission, this study we genotypically and pathotypically characterized 23 NDV isolates collected from chickens, ducks, pigeons live markets (LBMs) South part an H7N9 surveillance program during December 2013-February 2014. Abstract Newcastle disease virus (NDV) infection causes severe inflammation and is a highly contagious in poultry. Virulent NDV strains (GM) induce large quantities of interleukin-1β (IL-1β), which the central mediator inflammatory reaction. Excessive expression IL-1β exacerbates damage. Therefore, exploring mechanisms underlying NDV-induced can aid further understanding pathogenesis disease. Here, we showed that anti-IL-1β neutralizing antibody treatment decreased body temperature mortality...

Abstract Background Senecavirus A (SVA) caused porcine idiopathic vesicular disease (PIVD) showing worldwide spread with economic losses in swine industry. Although some progress has been made on host factors regulating the replication of SVA, role Z-DNA binding protein 1 (ZBP1) remains unclear. Methods The expression ZBP1 SVA-infected 3D/421 cells was analyzed by quantitative real-time PCR (qRT-PCR) and western blot. Western blot qRT-PCR were used to detect effects over interference SVA VP2...

In parts of southern China, some large‐scale swine farms are adjacent to lakes and ponds that home many types birds. Some will also raise poultry for consumption sale. Infection of host cells with the influenza virus is mediated by specific interactions between viral haemagglutinin (HA) and cell oligosaccharides containing sialic acid (SA) residues. Avian human viruses bind to alpha-2, 3 6 acid-linked receptors, respectively. To date, there have been no detailed tissue distribution data on receptors in domestic cat, a relatively new mammalian for infections. In this study, avian type was determined various organs (respiratory tract, gastrointestinal brain,...

From June to July 2017, six Seneca Valley virus (SVV) strains were isolated from swine herds exhibiting SVV-associated porcine idiopathic vesicular disease (PIVD) in Guangdong province, China. Complete genomic sequences of these newly identified genetically and phylogenetically analysed. The results revealed that SVV closely related USA/GBI29/2015 notably distinct all previous Chinese strains, indicating the reemergence new province.

Orf virus-encoded protein 002 (ORFV002) inhibits NF-κB signaling pathway by decreasing the acetylation of NF-κB-p65 through interference p65′s association with p300. However, precise mechanism how ORFV002 interferes p65/p300 is still unknown. Due to similarities amino acid sequences and adenovirus type 12 (Ad12) E1A (E1A-12), we hypothesized that N-terminal 52 acids might play an important role in this inhibition constructed several in-frame fusions enhanced green fluorescent (EGFP)...
