Carbohydrate derivatisation and glycocluster formation are both known to enhance avidity for lectin binding. Using a plant toxin human adhesion/growth-regulatory lectins (inter- intrafamily comparisons) the effect of their combination is examined. In detail, aromatic substituents were introduced at 2-N or 3′-positions N-acetyllactosamine products conjugated two types calix[n]arenes (n = 4, 6) viathiourea-linker chemistry.

Background Complement activation contributes to lung dysfunction in coronavirus disease 2019 (COVID-19). We assessed whether C5 blockade with eculizumab could improve outcome. Methods In this single-centre, academic, unblinded study two 900 mg doses were added-on standard therapy ten COVID-19 patients admitted from February 2020 April and receiving Continuous-Positive-Airway-Pressure (CPAP) ventilator support ≤24 hours. compared their outcomes those of 65 contemporary similar controls....

Cholera toxin (CT), the causative agent of cholera, displays a pentavalent binding domain that targets oligosaccharide ganglioside GM1 (GM1os) on periphery human abdominal epithelial cells. Here, we report first GM1os-based CT inhibitor matches valency (CTB). This contains five GM1os moieties linked to calix[5]arene scaffold. When evaluated by an inhibition assay, it achieved picomolar potency (IC50 = 450 pM) for CTB. represents significant multivalency effect, with relative inhibitory 100...

Nowadays, human oocytes/embryos are cryopreserved via slow freezing or vitrification. The aim of this study was to evaluate a rapid warming protocol for slow-frozen oocytes based on the standard procedure This prospective 216 sibling randomized either conventional thawing with vitrification solution. primary endpoint morphological assessment survival at 2h. Surviving were divided into two subgroups: (i) parthenogenetically activated; and (ii) fixed observed spindle/chromosome configuration....

Acute moderate to severe gastroenteritis is traditionally associated with hypernatremia but recent observations suggest that currently less common than hyponatremia. The latter has sometimes been documented also in children acute community-acquired diseases, such as bronchiolitis and pyelonephritis. We investigated the prevalence of dysnatremia gastroenteritis, This prospective observational study included 400 consecutive previously healthy infants ≥4 weeks ≤24 months age (232 males 168...

Four novel calix[4]arene-based glycoclusters were synthesized by conjugating the saccharide units to macrocyclic scaffold using CuAAC reaction and long hydrophilic ethylene glycol spacers. Initially, two galactosylcalix[4]arenes prepared starting from calixarene cores which differ in relative dispositions of alkyne azido groups. Once most convenient synthetic pathway was selected, further lactosylcalix[4]arenes obtained, one cone, other 1,3-alternate structure. Preliminary studies...

Levamisole is effectively used in steroid-dependent nephrotic syndrome and the more frequent side effects reported are cytopenia liver enzymes alterations. Several studies have demonstrated that this drug can induce high titers of circulating perinuclear antineutrophil cytoplasmic autoantibodies (ANCA) vasculitis, most them occurring case prolonged use. A four-year-old boy was affected with treated as a steroid-sparing agent at dose 2 mg/kg/48 h. After initiation treatment, number relapses...

Polyglycosylated calixarenes are efficient and selective multivalent ligands for lectins. However, the chemical decoration of these macrocyclic scaffolds with saccharides increasing complexity is hampered by highly complex chemistry carbohydrates. An alternative to conventional approach enzymatic diversification simple glycocluster-presented glycans. In this work, we present a chemo-enzymatic tetra-N-acetyl-lactosaminylcalix[4]arene via glycan extension catalyzed human...

Abstract Study question To date, no publications exist describing Blockchain in IVF (Hickman, 2020). Can technology be used for traceability and accountability laboratory procedures? Summary answer is an easy-to-implement incorruptible of a “Virus-Free” vitrification/warming procedure. What known already In healthcare, can become tool to address challenges regarding sensitive data-sharing medical procedures. Childhood-onset anti-neutrophil cytoplasmic antibody (ANCA)–associated vasculitis (AAVs) are a group of systemic autoimmune disorders characterized by inflammatory cells infiltration, necrosis small-medium blood vessels and autoantibodies to the neutrophil proteins leukocyte proteinase 3 (PR3-ANCA) or myeloperoxidase (MPO-ANCA). Primary AAVs in children rare with higher female preponderance, peak age at onset second decade median diagnosis 12–14 years [1]. They classified into [2]...
