Different structural changes of the Sym plasmid were found in a Rhizobium phaseoli strain that loses its symbiotic phenotype at high frequency. These rearrangements affected both nif genes and Tn5 mob insertions plasmid, some cases they modified expression bacterium's nodulation ability. One was more frequent heat-treated cells, but also under standard culture conditions; other appeared to be related conjugal transfer plasmid.

The symbiotic plasmid of Rhizobium leguminosarum biovar phaseoli strain CFN23 has been previously shown to undergo a series genetic rearrangements that modify the bacterial phenotype. A hyper-recombinant derivative was isolated. Summary Rhizobium leguminosarum biovar phaseol CFN23 loses its ability to nodulate beans at a high frequency because of deletion part symbiotic (pSym) plasmid (Soberón‐Chávez et al., 1986). We report here that least 80kb pSym are deleted upon loss the phenotype; removes nitrogenase structural nifHDK and common nod ABC genes. The size is not reduced, since it accompanied by an amplification other sequences. This genetic rearrangement similar yeast mitochondrial DNA leading to‘petite’mutations.
