Calcineurin, which binds to the Z-disc in cardiomyocytes via α-actinin, promotes cardiac hypertrophy response numerous pathologic stimuli. However, endogenous mechanisms regulating calcineurin activity muscle are not well understood. We demonstrate that a muscle-specific F-box protein called atrogin-1, or atrophy F-box, directly interacts with A and α-actinin-2 at of cardiomyocytes. Atrogin-1 associates Skp1, Cul1, Roc1 assemble an SCFatrogin-1 complex ubiquitin ligase activity. Expression...

Limitation of damage after ischemia and reperfusion injury to the myocardium remains an elusive clinical goal. Previous studies have suggested that molecular chaperones, which include members heat shock protein (Hsp) family, may cardioprotective effects, although protective role endogenous chaperones has not been well documented. CHIP (carboxyl terminus Hsp70-interacting protein) is a cochaperone/ubiquitin ligase integrates response stress at multiple levels. We tested CHIP(-/-) mice in vivo...

Endothelial cell migration is an important step during angiogenesis, and its dysregulation contributes to aberrant neovascularization. The bone morphogenetic proteins (BMPs) are potent stimulators of angiogenesis. Using microarray analyses, we find that myosin-X (Myo10) a BMP target gene. In endothelial cells, BMP6-induced Myo10 localizes in filopodia, BMP-dependent filopodial assembly decreases when expression reduced. Likewise, cellular alignment directional induced by BMP6 dependent....

Abstract Aim Characterize Growth Differentiation Factor 15 (GDF15) as a secreted biomarker of the integrated stress response (ISR) within central nervous system (CNS). Methods We determined GDF15 levels utilizing in vitro and vivo neuronal systems wherein ISR was activated. Primarily, we used murine model vanishing white matter disease (VWMD), neurological driven by persistent CNS, to establish link between cerebrospinal fluid (CSF) gene expression signature CNS. also CSF VWM patients....

Protein quality control and metabolic homeostasis are integral to maintaining cardiac function during stress; however, little is known about if or how these systems interact. Here we demonstrate that C terminus of HSC70-interacting protein (CHIP), a regulator control, influences the response pressure overload by direct regulation catalytic α subunit AMPK. Induction in Chip-/- mice resulted robust hypertrophy decreased energy generation stemming from failure activate Mechanistically, CHIP...

Purpose To evaluate the biodistribution, metabolism, and pharmacokinetics of a new type I collagen-targeted magnetic resonance (MR) probe, CM-101, to assess its ability help quantify liver fibrosis in animal models. Materials Methods Biodistribution, pharmacokinetics, stability CM-101 rats were measured with mass spectrometry. Bile duct-ligated (BDL) sham-treated imaged 19 days after procedure by using 1.5-T clinical MR imaging unit. Mice treated carbon tetrachloride (CCl4) or vehicle two...

Abstract Nicotinamide adenine dinucleotide (NAD) levels decline during aging, contributing to physical and metabolic dysfunction. The NADase CD38 plays a key role in age‐related NAD decline. Whether the inhibition of increases lifespan is not known. Here, we show that inhibitor 78c healthspan naturally aged mice. In addition 10% increase median survival, improved exercise performance, endurance, function effects were different between sexes. Our study first investigate effect animals.
