Atherosclerosis is a chronic inflammatory disease driven by hypercholesterolemia. During aging, T cells accumulate cholesterol, potentially affecting inflammation. However, the effect of cholesterol efflux pathways mediated ATP-binding cassette A1 and G1 (ABCA1/ABCG1) on cell-dependent age-related inflammation atherosclerosis remains poorly understood. In this study, we generate mice with cell-specific Abca1/Abcg1-deficiency low-density-lipoprotein-receptor deficient (Ldlr-/-) background....

Abstract Angiogenic sprouting relies on collective migration and coordinated rearrangements of endothelial leader follower cells. VE-cadherin-based adherens junctions have emerged as key cell-cell contacts that transmit forces between cells trigger signals during cell in angiogenesis. However, the underlying molecular mechanisms govern these processes their functional importance for vascular development still remain unknown. We previously showed F-BAR protein PACSIN2 is recruited to tensile...

BACKGROUND: Smooth muscle cells (SMCs) regulate blood flow distribution via vasoconstriction mediated by α-ARs (α-adrenergic receptors). Plasma membrane cholesterol accumulation affects α 1 -AR signaling and promotes loss of SMC contractile markers in vitro. ABCA1 ABCG1 (ATP-binding cassette transporter A1 G1) mediate efflux to HDL (high-density lipoprotein). ABCA1/ABCG1 show high expression medial low intimal SMCs atherosclerotic plaques. The role SMC-mediated atherogenesis remains poorly...

Background and aimsHigh lipoprotein (a) [Lp(a)] plasma levels are associated with increased atherosclerotic cardiovascular disease (ASCVD), in part attributed to elevated inflammation. High Lp(a) inversely correlate apolipoprotein [(APO(a)] isoform size. APO(a) size is negatively production rate (PR) positively fractional catabolic (FCR). We asked whether PR FCR (kinetics) of interleukin (IL)-6 IL-18, pro-inflammatory interleukins that promote ASCVD.MethodsWe used samples from existing data...

Glycogen storage disease type 1a (GSD Ia) is a rare inherited metabolic disorder caused by mutations in the glucose-6-phosphatase (G6PC1) gene. When untreated, GSD Ia leads to severe fasting-induced hypoglycemia. Although current intensive dietary management aims prevent hypoglycemia, patients still experience hypoglycemic events. Poor glycemic control associated with hypertriglyceridemia, hepatocellular adenoma and carcinoma, also an increased bleeding tendency of unknown origin. To...

Niemann-Pick type C1 (NPC1) disease is a progressive lysosomal storage disorder caused by mutations of the NPC1 gene. While neurodegeneration most severe symptom, large proportion patients also present with splenomegaly, which has been attributed to cholesterol and glycosphingolipid accumulation in late endosomes lysosomes. However, recent data reveal an increase inflammatory monocyte subset Npc1

The anti-inflammatory interleukin-1 receptor associated kinase-M (IRAK-M) is a negative regulator of MyD88/IRAK-4/IRAK-1 signaling. However, IRAK-M has also been reported to activate NF-κB through the MyD88/IRAK-4/IRAK-M myddosome in MEKK-3 dependent manner. Here we provide support that uses three surfaces its Death Domain (DD) downstream MyD88/IRAK-4/IRAK-M. Surface 1, with central residue Trp74, binds MyD88/IRAK-4. 2, Lys60, associates other DDs form an homotetramer under MyD88/IRAK-4...

Aging is associated with systemic, non-resolving inflammation and the accumulation of senescent cells. The resolution (or inflammation-resolution) in part mediated by balance between specialized pro-resolving mediators (SPMs) pro-inflammatory leukotrienes (LTs). Aged mice (i.e. 2 years age) exhibit a significant decrease SPM:LT ratio specific organs including spleen, which suggests that this organ may heightened be particularly amenable to SPM therapy. Previous studies have shown resolvin D1...

Glycogen storage disease type 1a (GSD Ia) is an inborn error of carbohydrate metabolism. Despite severe hyperlipidemia, GSD Ia patients show limited atherogenesis compared to age-and-gender matched controls. Employing a mouse model that resembles the hyperlipidemia in patients, we here found increased Ia. These data provide rationale for investigating larger patient cohort.
