Combined phototherapy and immunotherapy demonstrates strong potential in the treatment of metastatic cancers. An upconversion nanoparticle (UCNP) based antigen-capturing nanoplatform is designed to synergize phototherapies immunotherapy. In particular, this constructed via self-assembly DSPE-PEG-maleimide indocyanine green (ICG) onto UCNPs, followed by loading photosensitizer rose bengal (RB). ICG significantly enhances RB-based photodynamic therapy efficiency UCNP/ICG/RB-mal upon activation...

Chemical epigenetic manipulation of Penicillium citreonigrum led to profound changes in the secondary metabolite profile its guttate. While guttate from control cultures exhibited a relatively simple assemblage metabolites, collected treated with 50 μM 5-azacytidine (a DNA methyltransferase inhibitor) was highly enriched compounds representing at least three distinct biosynthetic families. The metabolites obtained fungus included six azaphilones (sclerotiorin (1), sclerotioramine (6),...

Background Physiological stress evokes rapid changes in both the innate and adaptive immune response. Immature αβ T cells developing thymus are particularly sensitive to stress, with infections and/or exposure lipopolysaccharide or glucocorticoids eliciting a apoptotic program. MicroRNAs class of small, non-coding RNAs that regulate global gene expression by targeting diverse mRNAs for degradation. We hypothesized subset thymically encoded microRNAs would be responsive modulate thymopoiesis....

TREX is a conserved multiprotein complex that necessary for efficient mRNA export to the cytoplasm. In Saccharomyces cerevisiae, additionally implicated in RNA quality control pathways, but it unclear whether this function mammalian cells. The Kaposi's sarcoma-associated herpesvirus (KSHV) ORF57 protein binds and recruits component REF/Aly viral mRNAs. Here, we demonstrate recruited KSHV noncoding polyadenylated nuclear (PAN) by ORF57. This recruitment correlates with ORF57-mediated...

Functional tuning of T cells based on their degree self-reactivity is established during positive selection in the thymus, although how differs for thymocytes with relatively low versus high unclear. In addition, preselection are highly sensitive to low-affinity ligands, but mechanism underlying enhanced cell receptor (TCR) sensitivity not fully understood. Here we show that murine experience briefer TCR signals and complete more slowly than those self-reactivity. Additionally, provide...

Abstract Background Metastatic breast cancer poses great challenge in treatment. N‐dihydrogalactochitosan (GC) is a novel immunoadjuvant that stimulates systemic immune responses when administered intratumourally following local tumour ablation. A combination of photothermal therapy (PTT) and GC, referred to as localized ablative immunotherapy (LAIT), extended animal survival generates an activated B cell phenotype MMTV‐PyMT mouse mammary microenvironment (TME). However, how T populations...

Rationale: B cells have emerged as key regulators in protective cancer immunity. However, the activation pathways induced during effective immunotherapy are not well understood. Methods: We used a novel localized ablative (LAIT), combining photothermal therapy (PTT) with intra-tumor delivery of immunostimulant N-dihydrogalactochitosan (GC), to treat mice bearing mouse mammary tumor virus-polyoma middle tumor-antigen (MMTV-PyMT).We single-cell RNA sequencing compare transcriptional changes by...

The tumor microenvironment (TME) promotes pro-tumor and anti-inflammatory metabolisms suppresses the host immune system. It prevents cells from fighting against cancer effectively, resulting in limited efficacy of many current treatment modalities. Different therapies aim to overcome immunosuppressive TME by combining various approaches synergize their effects for enhanced anti-tumor activity augmented stimulation Immunotherapy has become a major therapeutic strategy because it unleashes...

The thymus, an organ responsible for T cell development, is one of the more stress-sensitive tissues in body. Stress, form infections, radiation exposure, and steroids, impairs thymic epithelial (TEC) functions induces programmed death immature thymocytes. MicroRNAs are small noncoding RNAs involved tissue repair homeostasis, with several supporting development. We report that miR-205, epithelial-specific miR, maintains thymopoiesis following inflammatory perturbations. Thus, activation...

Autoreactive thymocytes are eliminated during negative selection in the thymus, a process important for establishing self-tolerance. Thymic phagocytes serve to remove dead thymocytes, but whether they play additional roles remains unclear. Here, using murine thymic slice model which undergo situ, we demonstrate that phagocytosis promotes selection, and provide evidence escape of autoreactive CD8 T cells periphery when thymus is impaired. We also show more efficient phagocyte presents...
