Purpose: To explore whether a cross-talk exists between PARP inhibition and PD-L1/PD-1 immune checkpoint axis, determine blockade of potentiates inhibitor (PARPi) in tumor suppression.Experimental Design: Breast cancer cell lines, xenograft tumors, syngeneic tumors treated with PARPi were assessed for PD-L1 expression by immunoblotting, IHC, FACS analyses. The phospho-kinase antibody array screen was used to the underlying mechanism PARPi-induced upregulation. therapeutic efficacy alone, or...
We have demonstrated the utility of hollow silica nanoparticles in fabricating conformal thin film nanoporous antireflection (AR) coatings on both poly(methyl methacrylate) (PMMA) and glass substrates. Layer-by-layer (LbL) assembly was successfully used to produce ultrathin AR planar textured surfaces. Hollow were synthesized extend range apparent refractive indices possible an coating, enabling design single index graded PMMA The diameter shell thickness are two independent, controllable...
Abstract In this study, thermosensitive amino‐silica@PDVB/PNIPAM Janus particles (JPs) are synthesized by seed emulsion polymerization‐induced phase separation and selective modification methods. Amino‐modified silica moieties covalently bonded to a diverse choice of substrates achieve robust composite coatings, PDVB/PNIPAM abdomen forms micro‐nano‐scale hierarchical surface. PNIPAM has lower critical solution temperature (LCST), which allows the hydrophilic hydrophobic properties coating...
Abstract The majority of breast cancer patients who achieve an initial therapeutic response to the HER2-targeted antibody trastuzumab will show disease progression within 1 year. Thus, identification novel agents that effectively inhibit survival cells have progressed on is critical. In current study, we dual epidermal growth factor receptor (EGFR)/human EGFR-2 (HER2) kinase inhibitor lapatinib induces apoptosis in trastuzumab-resistant derived from HER2-overexpressing SKBR3 line. Lapatinib...
Abstract The epidermal growth factor receptor (EGFR) has previously been detected in the nucleus of cancer cells and primary tumors. We have reported that EGFR translocates from plasma membrane to nucleus. Accumulation nuclear is linked increased DNA synthesis proliferation; however, pathological significance not completely understood. Here, we sought determine predictive value for survival ovarian patients, through examination 221 cases tissues by immunohistochemical analysis expression. In...
Accumulating evidence indicates that endocytosis plays an essential role in the nuclear transport of ErbB family members, such as epidermal growth factor receptor (EGFR) and ErbB-2. Nevertheless, how full-length receptors embedded endosomal membrane pass through pore complexes function non-membrane-bound nucleus remains unclear. Here we show upon EGF treatment, biotinylated cell surface EGFR is trafficked to inner (INM) complexes, remaining a membrane-bound environment. We further find...
Abstract Purpose: Results of multiple clinical trials suggest that EGF receptor (EGFR) tyrosine kinase inhibitors (TKI) exhibit negative effects on platinum-based chemotherapy in patients with lung cancer wild-type (WT) EGFR, but the underlying molecular mechanisms are still uncertain. Studies identify mechanism how TKIs negatively affect WT EGFR important for future development effective strategies to target cancer. Thus, we returned vitro study investigate and determine a possible...
Acetylation by ARD1 of an inhibitor mammalian target rapamycin may inhibit cancer growth.
Background/Aims: Autophagy is a dynamic catabolic process that maintains cellular homeostasis. Whether it plays role in promoting cell survival or death the of renal ischemia/reperfusion (I/R) remains controversial, partly because autophagy usually examined at certain time point. Therefore, monitoring whole course and apoptosis may help better understand I/R. Methods: were detected after mice subjected to bilateral ischemia followed by 0-h 7-day reperfusion, exposure TCMK-1 cells 24-h...
Abstract PARP1 inhibitors (PARPi) are currently used in the clinic for treatment of ovarian and breast cancers, yet their therapeutic efficacy against hepatocellular carcinoma (HCC) has been disappointing. To ensure PARPi HCC, a disease often diagnosed at intermediate to advanced stages with no effective options, it is critical identify not only biomarkers predict resistance but also rational treatments overcome this. Here, we report that heterodimer EGFR MET interacts phosphorylates Y907...
Drug resistance is a central challenge of cancer therapy that ultimately leads to treatment failure. In this study, we characterized mechanism drug arises AZD6244, an established mitogen-activated protein/extracellular signal-regulated kinase (MEK) 1/2 inhibitor currently being evaluated in clinical trials. AZD6244 enhanced the expression transcription factor FOXO3a, which suppressed cell proliferation. AZD6244-resistant cells, observed impaired nuclear localization reduced FOXO3a-mediated...
Abstract Objectives Tongue squamous cell carcinoma ( TSCC ) is the most frequent type of oral malignancy. Increasing evidence has shown that mi RNA s play key roles in many biological processes such as development, invasion, proliferation, differentiation, metabolism, apoptosis and migration. Materials methods qRT ‐ PCR analysis was performed to measure miR‐137 expression. CCK ‐8 analysis, colony formation, wound‐healing invasion were detect resultant functions. The direct target labelled...
The multifunctional gradient self-stratifying coating (SSC) is attractive in many applications, but the robustness and durability of functionalities still remains challenging. Here, a water-based SSC composed amphiphilic Janus nanoparticles (JNPs) binder latex constructed by an extremely simple one-step drying method. surface-partitioned "soft–hard" JNPs are specially designed synthesized, which hard side (fluorosilane modified silica) imparts hydrophobicity mechanical strength to SSC, soft...
Biotransformation of L-phenylalanine (L-Phe) to 2-phenylethanol (2-PE) was limited by product cytotoxicity. In order improve 2-PE production, a biotransformation strategy applying in situ adsorption investigated. Resin HZ818 chosen from several macroporous adsorbent resins, which adsorbed the most and least L-Phe. When 7% (dry w/v) resin added system, 6.6 g l−1 obtained 12 L-Phe, with molar yield 74.4%. Compared without addition resin, concentration improved 66.2%. Adsorption produced onto...
