Summary Chimeric antigen receptor T‐cell (CAR‐T) therapy has shown transformative potential in treating malignant tumours, with increasing global approval of CAR‐T products. However, high‐production costs and risks associated viral vector‐based cells—such as insertional mutagenesis secondary tumour formation—remain challenges. Our study introduces an innovative engineering approach using mRNA delivered via lipid nanoparticles (LNPs), aiming to reduce enhance safety while maintaining strong...

The reduction in insulin secretory capacity and beta-cell mass has been attributed, at least partially, to lipotoxicity, which may contribute the development of type 2 diabetes. Chronic free fatty acids (FFA) exposure impairs pancreatic function induces apoptosis. This study is elucidate underlying molecular mechanisms. We exposed INS-1E line palmitate or oleate, measured glucose stimulated secretion (GSIS). effect FFA on sterol regulatory element-binding protein (SREBP)-1c lipogenic pathway,...

The rapid development of quantitative portfolio optimization in financial engineering has produced promising results AI-based algorithmic trading strategies. However, the complexity markets poses challenges for comprehensive simulation due to various factors, such as abrupt transitions, unpredictable hidden causal and heavy tail properties. This paper aims address these by employing heavy-tailed preserving normalizing flows simulate high-dimensional joint probability complex environment...

Summary Type 1 diabetes (T1D) is due to a loss of immune tolerance islet antigens, such as glutamic acid decarboxylase 65 (GAD65), for which transplantation promising therapy. Therefore, the generation aiming at both alloantigen and GAD65 will help therapeutic intervention greatly in T1D. In this study, we tested effect programmed death-1 ligands (PD-L1)-transfected dendritic cells (DC) loaded with on alloresponse GAD65-reactive lymphocyte response. The DC2·4 cell line was transfected PD-L1...

Temporomandibular joint (TMJ) diseases are common clinical conditions. The number of patients with TMJ is large, and the etiology, epidemiology, disease spectrum, treatment remain controversial unknown. To understand master current situation occurrence, development prevention diseases, as well to identify patterns in incidence, drug sensitivity, prognosis crucial for alleviating patients'suffering. This will facilitate in-depth medical research, effective measures, formulation corresponding...

To explore whether T lymphocytes subgroup, B lymphocytes, platelet antigen CD41a, CD61 or antibodies are involved in the transfusion refractoriness.Forty-seven patients diagnosed as refractoriness and 32 that achieved effective therapy were ennolled this study. Flow cytometry was performed to detect proportion of cytotoxic cell (CD3(+)CD4(-)CD8(+)), helper (CD3(+)CD4(+)CD8(-)) (CD19 (+) ), expression glycoproteins, including CD41a CD61, while also measured by solid-phase agglutination.The...

Objective: To describe the clinical characteristics of sarcoidosis patients with arrhythmia as primary or main manifestation. Methods: We conducted a retrospective analysis arrhythmia-onset cases between January 2017 and December 2020. Their manifestations, radiological features, treatment prognosis were reviewed analyzed. Results: This study consisted 3 females 1 male, mean age 51 years (range from 42 to 58 old). Arrhythmia was first manifestation for all 4 cases, involving Ⅲ°...
