Abstract Ferroptosis plays a key role in the death of cells including cardiomyocytes, and it is related to variety cardiac diseases. However, ferroptosis‐related genes (FRGs) coronary artery disease (CAD) not well characterized. We downloaded CAD‐related information FRGs from gene expression omnibus (GEO) database Database (FerrDb) respectively. A total 10 DE‐FRGs were obtained, which closely linked autophagy regulation immune response. Subsequently, CA9 , CBS CEBPG HSPB1 SLC1A4 STMN1 TRIB3...
Vinblastine (VBL) has been considered as a first-line anti-tumor drug for many years. However, vinblastine-caused myocardial damage continually reported. The underlying molecular mechanism of the remains unknown. Here, we show that vinblastine induces and necroptosis is involved in vinblastine-induced both vitro vivo. results WST-8 flow cytometry analysis causes to H9c2 cells, animal experiments cell damage. necrosome components, receptor-interacting protein 1 (RIP1) 3 (RIP3), are...
The mortality of patients suffering from acute myocardial infarction is linearly related to the infarct size. As regeneration cardiomyocytes cardiac progenitor cells minimal in mammalian adult heart, we have explored a new therapeutic approach, which leverages capacity nanomaterials release chemicals over time promote protection and size reduction. Initial screening identified 2 chemicals, FGF1 CHIR99021 (a Wnt1 agonist/GSK-3β antagonist), synergistically enhance cardiomyocyte cell cycle...
Abstract 2’3’-cGAMP serves as a second messenger binding to the stimulator of interferon genes (STING) induce STING-dependent response, triggering downstream antiviral and anti-tumor mechanism. Ecto-nucleotide pyrophosphatase/phosphodiesterase-1 (ENPP1) hydrolyzes extracellular attenuates immune stimulatory tumor microenvironment, thereby facilitates progression. Inhibition ENPP1 activity in tumors that produce significant amount 2’3’-cGAMP, potentially activates response. We previously...
Right ventricle (RV) remodeling is a major pathological feature in pulmonary arterial hypertension (PAH). Magnesium lithospermate B (MLB) compound isolated from the roots of Salvia miltiorrhiza and it possesses multiple pharmacological activities such as anti-inflammation antioxidation. This study aims to investigate whether MLB able prevent RV PAH underlying mechanisms. In vivo, SD rats were exposed 10% O2 for 21 d induce remodeling, which showed hypertrophic features (increases ratio...
Anomalous origin of the right pulmonary artery from ascending aorta (AORPA) is a rare and potential fatal kind congenital heart disease. This study summarizes techniques outcomes 6 infants with AORPA who underwent surgical repair.Between November 2012 2014, received repair in Second Xiangya Hospital were included present study.Six (4 male, 66.7 %) median age 101.5 ± 70.0 days, body weight 4.13 0.62 kg at our institute. There no operative, in-hospital or follow-up deaths. Clinical symptoms...
The meager regenerative capacity of adult mammalian hearts appears to be driven by the proliferation endogenous cardiomyocytes; thus, strategies targeting mechanisms cardiomyocyte cell cycle regulation, such as Hippo/Yes-associated protein (Hippo/Yap) pathway, could lead development promising therapies for heart disease. pharmacological product TT-10 increases upregulating nuclear Yap levels. When intraperitoneal injections were administered infarcted mouse hearts, treatment promoted and was...
Hypericum perforatum [St. John's wort (SJW)] is known to cause a drug interaction with the substrates of cytochrome P450 (P450, CYP) isoforms, mainly CYP3A. This study aims determine dose response and time course effects SJW extract on P450s, UDP-glucuronosyltransferase (UGT), glutathione S-transferase (GST), NAD(P)H-quinone oxidoreductase (NQO) in mice. The oral administration male mice at 0.6 g/kg/d for 21 days increased hepatic oxidation activity toward Cyp3a substrate nifedipine. By...
Abstract Emerging studies revealed that tumor derived 2’3’-cGAMP could act as an immune transmitter and directly trigger anti-tumor immunity. Damage associated molecular patterns, e.g. damaged genomic DNA, released from injured cancer cells can be converted into 2’, 3’-cGAMP by cGAS. Subsequently, cGAMP triggers STING pathway activation to produce type I interferons (IFNs) other related innate immunity in the microenvironment. Ecto-nucleotide pyrophosphatase/phosphodiesterase-1 (ENPP1), a...
The actin-sequestering proteins, thymosin beta-4 (Tβ4) and hypoxia-inducible factor (HIF)-1α, are known to be associated with angiogenesis after myocardial infarction (MI). Herein, we aimed identify the mechanism of HIF-1α induction by Tβ4 investigate effects bone marrow mesenchymal stromal cells (BMMSCs) transfected gene (TMSB4) in a rat model MI.Rat BMMSCs were isolated, cultured, TMSB4 using lentivirus-mediated method. Rats surgically induced MI randomly divided into three groups (n =...
A 3-year-old Chinese boy was diagnosed with ipsilateral congenital malformations: right lung hypoplasia, dextroversion of heart, atrial septal defect, hepatic vein drainage directly into the atrium, facial asymmetry, microtia and deafness, indirect inguinal hernia. He underwent hernia repair at age 2. Although without any plastic surgery performed, he a defect recovered uneventfully. At 6-month follow-up, patient free from symptom dyspnea; his heart function returned to first grade.
Tetralogy of Fallot (TOF) with major aortopulmonary collaterals (MAPCA) is a well-known but always severe congenital heart disease. This study was designed to explore proper management after radical correction TOF MAPCA based on hierarchical approach. The following data were collected from 39 patients planned undergo TOF: age, weight, number collaterals, total lumen diameter and collateral diameter-to-body weight ratio, transcatheter occlusion cardiac catheterization findings, mechanical...
To introduce a new technique to create pulmonary valve biorifice for the reconstruction of right ventricular outflow tract in tetralogy Fallot (TOF), and summarize initial clinical experiment. The reconstructing with was used 53 cases TOF (the observation group). While conventional other 50 control The Data all reviewed retrospectively. The ages, weights, cardiopulmonary bypass time, cardiac arrest as well post operation ventilation support time were not different significantly between groups....
Abstract Cyclic GMP-AMP synthase (cGAS) recognizes and binds double-stranded DNA (dsDNA) in the cytosol to generate 2′3′-cGAMP, a cyclic dinucleotide an activator of STING-dependent innate immunity. Emerging studies revealed that tumor derived 2′3′-cGAMP could act as immune transmitter directly trigger anti-tumor Up date, Ecto-nucleotide pyrophosphatase/phosphodiesterase-1 (ENPP1/NPP1), membrane-bound nucleotide hydrolase, was discovered predominant enzyme hydrolyzing 2'3'cGAMP.
