Fully synthetic whole proteins in reach Solid-phase peptide synthesis of homogeneous peptides longer than about 50 amino acids has been a long-standing challenge because inefficient coupling and side reactions. Hartrampf et al. used an automated chemistry platform to optimize fast-flow were able produce fully single-domain (see the Perspective by Proulx). The targets included proinsulin enzymes such as barnase version HIV-1 protease containing multiple noncanonical acids. Refolded nearly...

Abstract Peptides play important roles in regulating biological processes and form the basis of a multiplicity therapeutic drugs. To date, only about 300 peptides human have confirmed bioactivity, although tens thousands been reported literature. The majority these are inactive degradation products endogenous proteins peptides, presenting needle-in-a-haystack problem identifying most promising candidate from large-scale peptidomics experiments to test for bioactivity. address this challenge,...

Peptide YY (PYY 3–36 ) has attracted attention in diabetes and obesity research because of its involvement food intake regulation glucose homeostasis. Native PYY maintains high potency on the Y 2 receptor with a loss 1 , 4 5 receptors. However, relatively short half-life, selectivity displayed by native peptide may not be optimal if long-acting analog is to developed. We performed variant screening identify key canonical amino acids that are pivotal selectivity, potency, stability. In...

Here we report highly efficient chemistry matched with an automated fast-flow instrument for the direct manufacturing peptide chains up to 164 amino acids over 328 consecutive reactions. The machine is rapid - chain elongation complete hours. We demonstrate utility this approach by chemical synthesis nine different protein that represent enzymes, structural units, and regulatory factors. After purification folding,...

DNA glue: A new alternate-strand linker (shown in green) has been synthesized that can connect two short sequences (5- and 9-mers, yellow) bind to double-stranded through major-groove interactions at pH 7.2 (see model). The resulting triple helices have high thermal stabilities as shown by melting temperatures exceeding 37 °C physiological values. The lack skills often makes it difficult make children young people experience the joy musical creativity, which essential element in education. This paper presents pilot project where modular robotics used create platform for creative expression that allows users genres without any prior knowledge or skills.
