Phagosomes are key organelles for the innate ability of macrophages to participate in tissue remodeling, clear apoptotic cells, and restrict spread intracellular pathogens. To understand functions phagosomes, we initiated systematic identification their proteins. Using a proteomic approach, identified >140 proteins associated with latex bead-containing phagosomes. Among these were hydrolases, proton pump ATPase subunits, fusion machinery, validating our approach. A series unexpected not...

We have examined the modifications occurring during transformation of phagosomes into phagolysosomes in J-774 macrophages. The use low density latex beads as markers (latex bead compartments, LBC) allowed isolation these organelles by flotation on a simple sucrose gradient. Two-dimensional gel electrophoresis, immunocytochemistry, and biochemical assays been used to characterize composition LBC at different time points after their formation, well interactions with endocytic pathway. Our...

The Nramp1 (natural-resistance-associated macrophage protein 1) locus (Bcg, Ity, Lsh) controls the innate resistance or susceptibility of mice to infection with a group unrelated intracellular parasites which includes Salmonella, Leishmania, and Mycobacterium. is expressed exclusively in professional phagocytes encodes an integral membrane that shares structural characteristics ion channels transporters. Its function mechanism action remain unknown. localization was analyzed control 129/sv...

Flotillin-1 was recently shown to be enriched on detergent-resistant domains of the plasma membrane called lipid rafts. These rafts, in sphingolipids and cholesterol, sequester certain proteins while excluding others. Lipid rafts have been implicated numerous cellular processes including signal transduction, trafficking, molecular sorting. In this study, we demonstrate both morphologically biochemically that are present phagosomes. structures flotillin-1 devoid main phagosomes protein...

The transformation of newly formed phagosomes into mature phagolysosomes is a process that involves complex series interactions between and other vacuolar organelles. machinery required by to mediate these poorly understood. In this study, we allowed human various rodent cells take up latex beads whose density facilitates simple purification using discontinuous sucrose gradients. With system, initiated systematic study phagosome proteins two-dimensional gel electrophoresis the currently...

Whereas amastigotes of the protozoan parasite Leishmania proliferate inside acidic phagolysosomal vacuoles macrophage, induced by donovani promastigotes during initiation infection are poorly characterized. Here, evidence is presented that interaction these parasitophorous with endocytic organelles very limited. In contrast, formed around L. mutants lacking cell surface lipophosphoglycan (LPG) fuse extensively endosomes and lysosomes. The role LPG repeating units in inhibition...

Highlights•The Leishmania metalloprotease GP63 cleaves phagosomal SNAREs, notably VAMP8•GP63-mediated VAMP8 cleavage inhibits phagolysosome biogenesis and cross-presentation•VAMP8 controls the proteolytic activity of phagosomesSummaryDuring phagocytosis, microorganisms are taken up by immune cells into phagosomes. Through membrane-trafficking events mediated SNARE proteins, phagosomes fuse with lysosomes, generating degradative phagolysosomes. Phagolysosomes contribute to host immunity...

ABSTRACT Phagolysosome biogenesis is essential for the killing and degradation of intracellular pathogens. It involves fusion phagosomes with various endocytic organelles, a process known to be regulated in part by Rab proteins. We generated RAW 264.7 macrophages expressing an active mutant Rab5 (Rab5(Q79L)) determine role phagocytosis phagolysosome biogenesis. Our results indicate that stimulates latex beads but not Fc or C3 receptor-mediated phagocytosis. also acts restrict complete...

ABSTRACT Among the small GTPases of Ras family, Rap proteins exhibit highest homology with p21Ras. The four so far identified constitute two subgroups, comprising Rap1(A,B) and Rap2(A,B) proteins. intracellular location Rap1A, Rap1B Rap2A was investigated in mammalian cells by confocal immunofluorescence microscopy. Using a specific anti-Rap1 affinity-purified antibody, both Rap1A were localized to late endocytic compartments (late endosomes/lysosomes) fibroblasts. localization B transiently...

We have shown recently that one of the survival strategies used by Leishmania donovani promastigotes during establishment infection in macrophages consists inhibiting phagosome-endosome fusion. This inhibition requires expression lipophosphoglycan (LPG), predominant surface glycoconjugate promastigotes, as parasites expressing truncated forms LPG reside phagosomes fuse extensively with endocytic organelles. In present study, we developed a single-organelle fluorescence analysis approach to...
