Narrow-band (312 nm) ultraviolet B light (UVB) is a new form of therapy for psoriasis, but its mechanism action unknown. In bilateral comparison clinical study, daily exposure psoriatic plaques to broad-band UVB (290–320 or 312-nm depleted T cells from the epidermis and dermis lesions. However, was significantly more depleting in both tissue compartments. To characterize cell depletion, assays apoptosis were performed on derived UVB-irradiated skin vivo irradiated vitro with UVB. Apoptosis...
To compare the therapeutic effectiveness of daily exposure to narrowband (NB) UV-B vs broadband (BB) with and without tar.Half-body exposures NB or BB were given for 4 weeks in this comparative treatment study. Narrowband was delivered from TL-01 fluorescent bulbs conventional same phototherapy cabinet. compared using a paired approach above waist tar (Goeckerman treatment) below waist.General clinical research center university hospital inpatient unit.Twenty-two patients moderate-to-severe...
Psoriasis is a chronic inflammatory skin disease in which epidermal hyperplasia results from infiltration by type I T lymphocytes and release of associated cytokines. A multifunctional cytokine, rhIL-11, modulates macrophage T-lymphocyte function cell culture shows anti-inflammatory activity animal models. We are testing subcutaneous delivery rhIL-11to patients with psoriasis phase 1 open-label dose-escalation clinical trial. Tissue was obtained lesional uninvolved before during treatment...
To determine whether 312-nm UV-B alters production of effector and regulatory cytokines by viable T cells that remain in psoriatic lesions during phototherapy.Prospective study.General clinical research center The Rockefeller University Hospital.Ten adult patients with moderate to severe psoriasis vulgaris was difficult manage were sequentially enrolled our protocols, biopsies taken at various time points from resolving lesions.Narrowband (312-nm) given starting 50% a minimum erythema dose,...
Abstract There is growing evidence that diesel exhaust particles (DEP) can induce allergic diseases with increased IgE production and preferential activation of Th2 cells. To clarify the cellular basis role DEP in induction Th2-dominant responses, we examined effects on cytokine by T cells stimulated anti-CD3/CD28 Ab monocyte-derived dendritic (MoDCs) CD40L and/or IFN-γ. We IFN-γ, IL-4, IL-5, IL-8, IL-10 produced TNF-α, IL-1β, IL-10, IL-12 MoDCs using real-time PCR analysis or ELISA....
<i>Background:</i> Munro’s microabscesses are a characteristic histopathologic feature of psoriasis vulgaris; however, the pathomechanisms underlying migration transepidermal leukocytes (PMNs) have not been fully elucidated yet. <i>Objective and Methods:</i> Since lesional scale extracts contain potent chemoattractants, such as IL-8 C5a fragments, we studied their location in lesions vulgaris PPP with immunohistochemical techniques. <i>Results:</i>...
Abstract Advances in the treatment of allergic disorders require elucidation autoregulatory immune systems induced averting detrimental inflammatory responses against invading foreign Ags. We previously reported that excessive Ags intruding through airway mucosa induce a subset regulatory CD4+ T cells secreting TGF-β regional mediastinal lymph nodes (MLNs), which inhibits Th2 and subsequent eosinophilic inflammation trachea. In present experiments we examined whether what mechanisms...
Abstract: Ber‐EP4 is an antibody to a cell membrane glycoprotein of unknown function. In the skin, immunoreactivity has been reported be localized in structures composed basaloid epithelial cells, i.e. fetal germ basal carcinoma, and trichoepithelioma as well eccrine or apocrine ducts. this study, we further characterized follicular expression at different stages hair cycle human terminal follicles. addition, clarify location + compared with keratin 15 19. Positive staining by was found...
An accurate method using image sharpness to determine the best focusing is proposed for ultra-high-voltage electron microscopy. This maximizes adjusting focus. Five images with different defocus values are used calculate sharpness. To obtain focus value that produces greatest sharpness, fitting quasi-Gaussian function five a suitable alternative. method, which gives better accuracy than wobbler microscope. The area can be selected without moving field of view, because at almost any in image.
We investigated the sequential changes in infiltrating inflammatory cells and several cytokine levels over a period of 48 h human back skin exposed to 3 minimal erythematous doses UVB. The measurement blood flow, using laser Doppler method, indicated that UVB-induced erythema reached peak 12-24 after irradiation. Immunohistochemically, an increase number CD4+ T was observed perivascular areas 6 UVB treatment continued for up h. CD8+ were scarce until 24 h, but their numbers gradually...
Abstract The reaction of 6-amino-6-deoxycelluloe (1) with hematin in the presence N,N′-carbonyldiimidazole and 1,8-diazabicyclo[5.4.0]undec-7-ene N,N-dimethylformamide at 50°C for four days afforded hematin-appended 6-aminocellulose (2). Compound 2 was more stable than hematin, retained oxidation activity even after being treated 100°C 3 h. showed good guaiacol an aqueous 80% AcOH solution (pH 1.5), whereas horseradish peroxidase very low activities under same conditions, suggesting that...
Abstract We report a 9‐year‐old Japanese female patient with atopic dermatitis associated idiopathic thrombocytopenic purpura. She demonstrated high serum immunoglobulin (Ig)E and IgE specific to several environmental allergens, but extremely low thymus activation‐regulated chemokine ( TARC ) levels regardless of the disease progression. This case suggests platelets as main source .
