There are numerous reports about particle–cell interaction studies in the literature. Many of those performed two-dimensional cell cultures. While interpretation such seems trivial at first sight, fact for quantitative analysis some basic physical and physicochemical bases need to be considered. This starts with dispersion particles, which gravity, Brownian motion, interparticle interactions The respective strength these determines whether particles will sediment, dispersed, or agglomerated....
Self-assembling peptide hydrogels with faster gelation kinetics and higher mechanical rigidity are favorable for their practical applications. A design strategy to control the folding, self-assembly, hydrogelation of β-hairpin peptides via hydrophobic amino acid substitutions has been explored in this study. Isoleucine hydrophobicity stronger propensity β-sheet hydrogen bonding than valine. After valine residues MAX1 (VKVKVKVKVDPPTKVKVKVKV-NH2) were replaced isoleucines, oscillatory...
Highly fluorescent carbon dots (CDs) with quantum yields up to 96% were rapidly synthesized within 5 min via microwave irradiation controllable temperature. Multifunctional bioimaging including live cell staining, cell-specific targeting and in vivo imaging further demonstrated by using high quality low cost CDs as contrast agents.
Selective tumour cell imaging and synergistic anti-cancer therapeutics are achieved by using the conjugate of AS1411 graphene quantum dots.
Using on-line microdialysis, we have characterized in vivo dynamics of pineal 5-hydroxytryptamine (5-HT; serotonin) release. Daily 5-HT output is triphasic: ( i ) levels are constant and high during the day; ii early night, there a novel sharp rise synthesis release, which precedes nocturnal melatonin synthesis; iii late low. This triphasic production persists darkness influenced strongly by intrusion light at night. We demonstrate that both diurnal release activated sympathetic innervation...
Abstract: Serotonin (5‐hydroxytryptamine, 5‐HT), a precursor for melatonin production, is produced abundantly in the pineal gland of all vertebrate animals. The synthesis 5‐HT rate limited by tryptophan hydroxylase 1 (TPH1) whose activity displays twofold increase at night. Earlier studies from our laboratory demonstrate that secretion exhibits dynamic circadian rhythms with elevated levels during early night, and controlled adrenergic signaling In this study, we report (a) total output TPH1...
The nucleolus is an important subnuclear structure and there are very few dyes available in the market for nucleolar imaging. Subcellular organelles delivery presents a common stumbling block many nanomaterial-based applications, including intracellular fluorescence staining. We now introduce novel luminescent graphene quantum dot (nGQD), which able to selectively light up nucleoli of living cells, address these challenges. Investigations on subcellular localization different GQDs have...
Mesenchymal stromal cells (MSCs) have an inherent migratory capacity towards tumor tissue in vivo. With the future objective to quantify homing efficacy of MSCs, as first step this direction we investigated use inorganic nanoparticles (NPs), particular ca. 4 nm-sized Au NPs, for MSC labeling. Time dependent uptake efficiencies NPs at different exposure concentrations and times were determined via inductively coupled plasma mass spectrometry (ICP-MS). The labeling efficiency MSCs was terms...
Novel active tumor targeting fluorescent gold nanoclusters are synthesized through a facile method.
Exhaust emissions from vessels have increasingly attracted attention in the continuously growing marine transport world trade market. The International Maritime Organization (IMO) has introduced a number of measures designed to reduce exhaust global shipping. As one busiest ports world, Qingdao port been studied propose possible support development efficient emission reduction. In this study, large amount data inventory was used predict its annual emissions, and hence, help understand...
Functional studies support the presence of Na + -HCO 3 − cotransporter (NBC) in corneal endothelium and possibly epithelium; however, molecular identification membrane localization have not been reported. To test whether NBC is expressed bovine cornea, Western blotting was performed, which showed a single band at ∼130 kDa for freshly isolated cultured endothelial cells, but no epithelium. Two isoforms recently cloned kidney (kNBC) pancreas (pNBC). RT-PCR run using fresh epithelial total RNA...
HCO[Formula: see text]-dependent fluid secretion by the corneal endothelium controls hydration and maintains transparency. Recently, it has been shown that mRNA for cystic fibrosis transmembrane conductance regulator (CFTR) is expressed in endothelium; however, protein expression, functional localization, a possible role text]transport have not reported. Immunoblotting CFTR showed single band at ∼170 kDa both freshly isolated primary cultures of bovine endothelial cells. Indirect...
Purpose. Chloride and bicarbonate are necessary for maintenance of fluid transport by the corneal endothelium, however there is little information on identity anion proteins that could serve as efflux mechanisms in endothelial cells. Therefore, we ask whether mRNA related proteins, CFTR, CLC-2, ClC-3, ClC-5 AE2, expressed human, bovine or rabbit endothelium. Methods. RT-PCR was performed AE2 using total RNA from fresh endothelium well cultured cells (CBCEC). Specificity PCR products...
Corneal transparency and hydration control are dependent on HCO(3)(-) transport properties of the corneal endothelium. Recent work (13) suggested presence an apical 1Na(+)-3HCO(3)(-) cotransporter (NBC1) in addition to a basolateral 1Na(+)-2HCO(3)(-) cotransporter. We examined whether NBC1 contributes significantly or permeability participates transendothelial net flux cultured bovine protein expression was reduced using small interfering RNA (siRNA). Immunoblot analysis showed that 5-15 nM...
The 3-O-sulfotransferases (3OSTs) catalyze the addition of sulfate groups at 3-OH site glucosamine in heparan proteoglycans, which serve as critical mediators various biological functions. We demonstrate that 3OST2 isoform is expressed high levels rat pineal specifically during daylight hours. dramatic diurnal rhythm regulated by central clock-controlled activities superior cervical ganglion, persists constant darkness, and inducible light nighttime. Importantly, transcription blocked...
Carbonic anhydrase activity has a central role in corneal endothelial function. The authors examined the of carbonic IV (CAIV) facilitating CO(2) flux, HCO(3)(-) permeability, and flux across apical membrane.Primary cultures bovine cells were established on membrane-permeable filters. Apical CAIV was inhibited by benzolamide or siRNA knockdown CAIV. fluxes permeability determined measuring pH(i) changes response to altering gradient membrane. Basolateral (B-to-A) pH weakly buffered bath...

Abstract: This study describes the development of a new technique for long‐term measurement daily 5‐hydroxytryptamine (5‐HT) and melatonin contents in pineal gland freely moving rats. The features number novel improvements over previous protocols. It allows visualization accurate targeting guide cannula, which minimizes bleeding; incurs no direct injury to surrounding brain tissues; causes interference with sympathetic innervation from superior cervical ganglia. Robust releases indole...
