Regulatory T (T reg) cells exert powerful down-modulatory effects on immune responses, but it is not known how they act in vivo. Using intravital two-photon laser scanning microscopy we determined that, the absence of reg cells, locomotion autoantigen-specific inside lymph nodes decreased, and contacts between antigen-loaded dendritic (DCs) are longer duration. Thus, can an early effect responses by attenuating establishment stable during priming naive DCs.

Implantation of the fertilized egg into maternal uterus depends on fine balance between inflammatory and anti-inflammatory processes. Whilst regulatory T cells (Tregs) are reportedly involved in protection allogeneic fetuses against rejection by immune system, their role for pregnancy to establish, e.g., blastocyst implantation, is not clear. By using 2-photon imaging we show that Foxp3(+) accumulated mouse during receptive phase estrus cycle. Seminal fluid further fostered Treg expansion....

A complete understanding of the mechanisms underlying acquisition protective immunity is crucial to improve vaccine strategies eradicate malaria. However, it still unclear whether recognition damage signals influences immune response Plasmodium infection. Adenosine triphosphate (ATP) accumulates in infected erythrocytes and released into extracellular milieu through ion channels erythrocyte membrane or upon rupture. The P2X7 receptor senses ATP induces CD4 T cell activation death. Here we...

Dendritic cells (DCs) are phagocytes that highly specialized for antigen presentation. Heterogeneous populations of macrophages and DCs form a phagocyte network inside the red pulp (RP) spleen, which is major site control blood-borne infections such as malaria. However, dynamics splenic during Plasmodium poorly understood, limiting our knowledge regarding their protective role in Here, we used vivo experimental approaches enabled us to deplete or visualize order clarify these issues. Dendritic cells (DCs) hold promise for anti-cancer immunotherapy. However, clinically, their efficiency is limited and novel strategies to improve DC-mediated anti-tumor responses are needed. Human DCs display high content of sialic acids, which inhibits maturation co-stimulation capacity. Here, we aimed understand whether exogenous desialylation improves immunity. Compared fully sialylated DCs, desialylated human loaded with tumor-antigens showed enhanced ability induce autologous T...

Malaria in pregnancy is exquisitely aggressive, causing a range of adverse maternal and fetal outcomes prominently linked to Plasmodium-infected erythrocyte cytoadherence trophoblast. To elucidate the physiopathology infected erythrocytes (IE) sequestration placenta we devised an experimental system for intravital placental examination P. berghei-infected mice. BALB/c females were mated C57Bl/6 CFP+ male mice with GFP+ berghei IE, at gestational day 18, placentas exposed time-lapse imaging...

Patients infected by Leishmania braziliensis develop debilitating skin lesions. The role of inhibitory checkpoint receptors (ICRs) that induce T cell exhaustion during this disease is not known. Transcriptional profiling identified increased expression ICRs including PD-1, PDL-1, PDL-2, TIM-3, and CTLA-4 in lesions patients was confirmed immunohistology where there both CD4 + CD8 subsets. Moreover, PDL-1/PDL-2 ligands were on macrophages compared to healthy controls. proportions PD1 , but...

Trypanosoma cruzi infection in mice is associated with severe hematological changes, including anemia, which may contribute to mortality. TNF-α and nitric oxide (NO) play a critical role establishing host resistance this pathogen. We hypothesized that phagocyte-derived NO damages erythrocytes contributes the anemia observed during T. infection. To test hypothesis, two strains of differed susceptibility response were used these studies. also blocked endogenous production by aminoguanidine...

Abstract Recent studies have revealed an important role for CTLA-4 as a negative regulator of T cell activation. In the present study, we evaluated importance to immune response against intracellular protozoan, Trypanosoma cruzi, causative agent Chagas’ disease. We observed that expression in spleen cells from naive mice cultured presence live trypomastigote forms T. cruzi increases over time exposure. Furthermore, harvested recently infected showed significant increase when compared with...

It is postulated that accumulation of malaria-infected Red Blood Cells (iRBCs) in the liver could be a parasitic escape mechanism against full destruction by host immune system. Therefore, we evaluated vivo this and its potential immunological consequences. A massive P. c. chabaudi AS-iRBCs (Pc-iRBCs) was observed intravital microscopy along with an over expression ICAM-1 on day 7 infection, as measured qRT-PCR. Phenotypic changes were also regulatory T cells (Tregs) dendritic (DCs) isolated...

Analysis spleen reveals most LOW TCR cells have an experienced phenotype (CD44 HIGH CD62L CD45RA,B,C ). These small size, lack activation markers such as CD69, CD25 CD11b (Mac‐1), do spontaneously secrete cytokines, suggesting they are at resting state. When...
