Summary: A multicenter, double‐blind, randomized, placebo‐controlled study evaluated the efficacy and safety of gabapentin (Neurontin, GBP) as add‐on therapy in 272 patients with refractory partial seizures who were receiving one to two standard antiepileptic drugs (AEDs). Efficacy assessments compared frequency during 12‐week treatment phase (T) baseline period (B). The primary analysis data for GBP 900 mg/day placebo; 1,200‐mglday group provided doseresponse data. criteria percentage...

Members of the cytochrome P450 (P450) family drug-metabolizing enzymes are present in human brain, and they may have important roles oxidation endogenous substrates. The polymorphic CYP2D6 is one major brain isoforms has been implicated neurodegeneration, psychosis, schizophrenia, personality traits. objective this study was to determine whether endocannabinoid arachidonoylethanolamide (anandamide) a substrate for CYP2D6. Anandamide ligand cannabinoid receptor CB1, which also activated by...

Summary: Purpose: Gabapentin (GBP) is a commonly used drug in the treatment of partial seizures, but its mode action still unclear. The genesis seizures temporal lobe epilepsy thought to be crucially influenced by intrinsic membrane properties. Because I h substantially contributes properties neurons, effects GBP on were investigated CA1 pyramidal cells rat hippocampus. Methods: hippocampal slices examined using whole‐cell patch‐clamp technique. Results: increased amplitude...

Abstract The hyperpolarization‐activated current ( I h ) contributes to intrinsic properties and network responses of neurons. Its biophysical depend on the expression profiles underlying hyperpolarization‐activated, cyclic nucleotide‐gated (HCN) channels presence AMP (cAMP) that potently differentially modulates conducted by HCN1, HCN2 and/or HCN4. Here, we studied in hippocampal CA1 pyramidal cells, developmental evolution HCN‐subunit isoforms contribute this current, their interplay with...

As one of the best studied photoswitches, spiropyrans (SPs) have attracted significant interest in scientific community. Among many stimuli to alter isomerization into merocyanine (MC) isomer, such as temperature, pH, solvent polarity, redox potential, or mechanical force, ability MC form act a ligand site for metal complexation has recently raised new attention. We herein synthesize hitherto undescribed coordination compounds...

Summary: Purpose: To report on the occurrence of myoclonus in patients receiving pregabalin (PGB) for treatment focal epilepsy. Methods: Clinic records 19 who were consecutively enrolled at a tertial referral epilepsy center randomized, double‐blind and/or open add‐on study with PGB reviewed. Results: In four treated PGB, newly developed. The side effect appeared doses 50–600 mg/day; intensity showed some dose dependency. All had medically refractory and received other antiepileptic drugs...

In the presence of silver(I) fluoride, highly fluorinated olefins react readily under solvent-free conditions with arenes via CH-substitution. This transformation could be used to synthesize various methoxycarbonyltetrafluoroethylated aromatic triazenes and anisoles high functional group tolerance. The method applied synthesis a formal bioisostere NSAID flurbiprofen. To best our knowledge, this is first example which uses for perfluoroalkylation substrates.
