Sodium/proton exchanger 1 (NHE1) is an electroneutral secondary active transporter present on the plasma membrane of most mammalian cells and plays critical roles in regulating intracellular pH volume homeostasis. Calcineurin B-homologous protein (CHP1) obligate binding partner that promotes NHE1 biosynthetic maturation, cell surface expression pH-sensitivity. Dysfunctions either are associated with neurological disorders. Here, we elucidate structures human NHE1-CHP1 complex both inward-...

Bicarbonate facilitates mucin unpacking and bacterial killing; however, its transport mechanisms in the airways are not well understood. cAMP stimulates anion efflux through cystic fibrosis (CF) transmembrane conductance regulator (CFTR; ABCC7) channel, this is defective CF. The exchanger pendrin (SLC26A4) also mediates HCO3- upregulated by proinflammatory cytokines. Here, we examined CFTR expression their contributions to secretion human nasal bronchial epithelia. In native tissue, both...

CFTR (cystic fibrosis transmembrane conductance regulator) is a tightly regulated anion channel that mediates chloride and bicarbonate in many epithelia other tissues, but whether its regulation varies depending on the cell type has not been investigated. Epithelial expression highest rare cells called ionocytes. We studied control ionocyte-enriched cultures by transducing bronchial basal with adenoviruses encode only eGFP or FOXI1 (forkhead box I1) + as separate polypeptides. dramatically...

The airway mucosal microenvironment is crucial for host defense against inhaled pathogens but remains poorly understood. We report here that the surface normally undergoes surprisingly large excursions in pH during breathing can reach 9.0 inhalation, making it most alkaline fluid body. Transient alkalinization requires luminal bicarbonate and membrane-bound carbonic anhydrase 12 (CA12) antimicrobial. Luminal concentration CA12 expression are both reduced cystic fibrosis (CF), mucus...

The pH of airway epithelial secretions influences bacterial killing and mucus properties is reduced by acidic pollutants, gastric reflux, respiratory diseases such as cystic fibrosis (CF). effect acute acid loads depends on buffer capacity, however the buffering has not been well characterized. In this work we develop a method for titrating micro-scale (30 μl) volumes use it to study fluid secreted human cell line Calu-3, widely used model submucosal gland serous cells. Microtitration curves...

The cystic fibrosis transmembrane conductance regulator (CFTR) is a tightly regulated anion channel that mediates secretion by epithelia and mutated in the disease fibrosis. CFTR forms macromolecular complexes with many proteins; however, little known regarding its associations membrane lipids or regulation of distribution mobility at cell surface. We report here secretagogues (agonists stimulate secretion) such as peptide hormone vasoactive intestinal (VIP) muscarinic agonist carbachol...

Anion exchanger type 2 (AE2 or SLC4A2) is an electroneutral Cl(-)/HCO(3)(-) expressed at the basolateral membrane of many epithelia. It thought to participate in fluid secretion by airway However, role AE2 remains uncertain, due lack specific pharmacological inhibitors, and because it electrically silent therefore does not contribute directly short-circuit current (I(sc)). We have studied Cl(-) epithelial cell line Calu-3. After confirming expression its mRNA protein, a knock-down called...

Bicarbonate plays an important role in airway host defense, however, its transport mechanisms remain uncertain. Here we examined the relative contributions of anion channel CFTR (cystic fibrosis transmembrane conductance regulator, ABCC7) and exchanger pendrin (SLC26A4) to HCO3− secretion by human cell line Calu-3. Pendrin were both detected parental Calu-3 cells, although mRNA protein expression appeared higher for than pendrin. Targeting transcripts with lentiviral shRNA reduced detection...

<p class="Abstract">We have previously identified an antimicrobial peptide called dybowskin2-CDYa (Dy2) in the cutaneous secretion from Chinese frog Rana dybowskii. In this study, we used Dy2 as a template to prepare some novel peptides with improved stability and hydrophobicity. The activities exerted by these against Gram positive negative bacteria were evaluated MIC CFU assays under physiological conditions. One peptide, AWRK6, was 2-4 fold more potent than Dy2, toxic most of...

The number of...
