Neonatal sepsis is a major cause of mortality in preterm infants, with Escherichia coli as one the leading pathogens. Few studies have examined interplay between virulence factors, resistance profiles, phylogroups, and clinical outcomes this population. We analyzed 52 E. strains isolated from 49 neonates diagnosed at tertiary-level hospital Mexico. Strains underwent phylogenetic classification, gene profiling, antimicrobial testing. PFGE was used to assess genetic relatedness outbreak...
Urinary tract infections (UTIs) are a leading cause of illness in children and adults all ages, with uropathogenic Escherichia coli (UPEC) being the primary agent responsible. During colonization subsequent infection urinary (UT), UPEC requires expression genes associated virulence, such as those that encode fimbrial adhesins FimH, PapG, CsgA, well presence TosA protein flagellar appendages bacteria. However, for to be successful, must overcome host’s immunological barriers, physical...
Abstract Background Nontuberculous mycobacteria (NTM) are environmental opportunistic pathogens found in natural and human-engineered waters, including drinking water distribution systems household plumbing. This pilot study examined the frequency of occurrence NTM potable samples Mexico City. Potable were collected from “main house faucet” kitchen faucet. The presence aerobic-mesophilic bacteria (AMB), total coliforms (TC), fecal (FC) species determined. Mycobacteria identified by PCR...
The aims of this study were to evaluate the microbiological quality and occurrence nontuberculous mycobacteria (NTM) in a variety salads sprouts from supermarkets street vendors Mexico City. Aerobic-mesophilic bacteria (AMB) present 100% RTE-salads samples; 59% samples outside guidelines range (>5.17 log10 CFU per g). Although fecal coliforms (FC) 32% samples, only 8% them exceeded permissible limit (100 MPN/g). Regarding 100 RTE-sprouts, all also positive for AMB total (TC) 69% FC. Seven...
Nontuberculous mycobacteria (NTM) have recently been recognized as important species that cause disease even in immunocompetent individuals. The mechanisms these use to infect and persist inside macrophages are not well characterised. To gain insight concerning this process we used THP-1 infected with M. abscessus , fortuitum celatum tuberculosis . Our results showed slow-growing gained entrance into cells more efficiency than fast-growing mycobacteria. We also demonstrated viable persisted...
Helicobacter pylori is a major aetiologic agent associated with gastritis. H. infections increase the expression of Toll-like receptor (TLR), which in turn modulates microRNA (miRNA)-146a and miRNA-155. The objective this study was to compare miRNA-146a miRNA-155 gastric lesions paediatric adult patients different pathologies Mongolian gerbils (Meriones unguiculatus) infected 26,695.Quantification miRNA performed by quantitative real-time polymerase chain reaction (qRT-PCR) paraffin-embedded...
Abstract Background The prevalence of infections with Mycobacterium tuberculosis (MTb) and nontuberculous mycobacteria (NTM) species in HIV-infected patients Mexico is unknown. aims this study were to determine the frequency MTb NTM from City, evaluate genotypic diversity complex strains, their drug resistance profiles by colorimetric microplate Alamar Blue assay (MABA), finally, detect mutations present kat G, rpo B inh A genes, resulting isoniazid (INH) rifampin (RIF) resistance. Results...
ABSTRACT Mycobacteria are thought to have either one or two rRNA operons per genome. All mycobacteria investigated date an operon, designated rrnA , located downstream from the murA gene. We report that Mycobacteriun fortuitum has a second rrn rrnB which is tyrS gene; very close 3" end of gene ( 3-mag ) coding for 3-methylpurine-DNA-glycosylase. The operon Mycobacterium smegmatis was shown similar organization, namely, 5" - 3". M. found single dedicated promoter. During exponential growth in...
Background Mexico is one of the most important contributors drug and multidrug-resistant tuberculosis in Latin America; however, knowledge genetic diversity drug-resistant isolates limited. Methods In this study, structure 112 Mycobacterium strains from southeastern was determined by spoligotyping 24-loci MIRU-VNTRs. Findings The results show eight major lineages, which T1 (24%), followed LAM (16%) H (15%). A total 29 (25%) were identified as orphan. abundant SITs SIT53/T1 SIT42/LAM9 with 10...
Bovine tuberculosis is a zoonotic disease that not only causes huge economic losses but also poses an important risk for human infection. The definitive identification of clinical isolate relies on time-consuming, highly specialized and laborious biochemical tests. We have developed method the rapid reliable Mycobacterium bovis its simultaneous differentiation from other members M. complex. Furthermore, technique allowed us to distinguish complex Mycobacterial species. comprises both single...
The ParA/Soj and ParB/Spo0J proteins, the cis-acting parS site, participate actively in chromosome segregation cell cycle progression. Genes homologous to parA parB, two putative copies, have been identified Mycobacterium bovis BCG smegmatis chromosomes. As tuberculosis, parB genes these non-pathogenic mycobacteria are located near chromosomal origin of replication. present work focused on determination transcriptional organisation ~6 Kb orf60K-parB region M. by primer extension, fusions...
Klebsiella oxytoca is a resident of the human gut. However, certain K. toxigenic strains exist that secrete nonribosomal peptide tilivalline (TV) cytotoxin. TV pyrrolobenzodiazepine causes antibiotic-associated hemorrhagic colitis (AAHC). The biosynthesis driven by enzymes encoded aroX and NRPS operons. In this study, we determined effect environmental signals such as carbon sources, osmolarity, divalent cations on transcription both biosynthetic Gene expression was enhanced when bacteria...
Background: Drug-resistant tuberculosis (TB) is associated with higher mortality rates in patients human immunodeficiency virus (HIV). In Mexico, the number of deaths due to TB among HIV-positive population has tripled recent years. Methods: Ninety-three Mycobacterium strains isolated from same HIV-infected treated a public hospital Mexico City were studied determine drug resistance first- and second-line anti-TB drugs identify mutations resistance. Results: Of 93 patients, 82.7% new cases,...
This study aimed to determine the bacteriological quality and presence of diarrheagenic Escherichia coli pathotypes (DEP) nontuberculous mycobacteria (NTM) species in 85 packaged ice samples from 12 different states central Mexico. Three had a pH 9.8 therefore fell outside acceptable range for pH. All were positive aerobic-mesophilic bacteria, with limits ranging 1 3.47 log CFU/mL. In total, 35, 11, 3 total coliforms (TC), fecal (FC), E. coli, respectively. samples, TC concentration ranged...
The general features of the shift to a dormant state in mycobacterial species include several phenotypic changes, reduced metabolic activities, and increased resistance host environmental stress conditions. In this study, we aimed provide novel insights into viability morphological changes M. smegmatis that contribute its long-term survival under starvation or hypoxia. To end, conducted assays evaluate cell viability, gene expression. During starvation, exhibited reduction length, presence...
SPI-2 encodes a type III secretion system (T3SS) that is hallmark for the species Salmonella enterica , which essential survival and replication within macrophages. Expression of genes positively controlled by two-component SsrAB.
Cholesterol has been reported to play an important role during Mycobacterium tuberculosis infection and its dormant state inside the host. We present determination of proteomic profiles M. H37Rv in presence cholesterol as sole carbon source under exponential growth two vitro dormancy phases (NRP1 NRP2). Using 2D-PAGE, we detected that expressed a high diversity proteins both non-replicative phases. also found was involved overexpression some related sulfur metabolism (CysA2), electron...
