To analyze the structure of LH in three patients with immunologically anomalous LH, whole coding region beta-subunit gene was examined. These were infertile, and their serum levels could not be measured an immunoassay kit. Immunoblotting showed no marked changes molecular size beta. Genomic DNA extracted from peripheral lymphocytes normal controls, beta genes amplified by polymerase chain reaction technique, using primer pairs that are capable specifically amplifying only without...

For women with congenital uterine infertility, or for those who have undergone hysterectomy, transplantation is one of the potential treatments to regain fertility. In this study, we utilized a primate model transplantation, and evaluated patency our microsurgical anastomoses, perfusion transplanted uterus. Two female cynomolgus monkeys underwent surgery. We anastomosed two arteries vein in Case 1 veins 2. The used were anastomosis was done external iliac artery. ovarian both animals, but...

Abstract Aim Uterine transplantation ( UTx ) is a potential option for child‐bearing in women with uterine infertility. Recovery of function after allogeneic non‐human primates has not been reported. The objective this study to establish the functional transplant model primates. Methods Uteri two cynomolgus monkeys were simultaneously removed, cooled at 4° C and perfused heparin saline. uteri interchanged each other then orthotopically transplanted. BACKGROUNDUterus transplantation may be the only theoretical option for some women, example, those with congenital uterine infertility or who have undergone hysterectomy. In this study, we evaluated intra- and post-operative blood flow conditions of vascular anastomosed regions blood-perfused area transplanted uterus in a cynomolgus macaque model autotransplantation.

To study the aging process in human ovary, we analyzed accumulation of mitochondrial DNA (mtDNA) mutation ovary women various ages. Thirty-nine tissues from ovaries and abdominal muscles, ranging those a fetus to 77-yr-old woman, were collected at artificial abortion or gynecological surgery. Total was extracted these tissues, parts mtDNA amplified via polymerase chain reaction. Through experiments, found 4977-bp deletion between ATPase8 ND5 genes ovarian samples obtained menopausal...

To clarify whether apoptosis is involved in endometriosis, we obtained eutopic endometrial tissues along with endometriotic from the uterus (adenomyosis) (n = 12) and ovary patients undergoing gynaecological surgery. Apoptosis-induced DNA fragmentation was detected by TdT-mediated dUTP-biotin nick-end labelling method, immunostaining a monoclonal antibody against Fas, Ley or B-cell leukaemia/lymphoma-2 (bcl-2) also performed using same tissue section. Analysis showed that occurring all...

Abstract Previous kinetic studies have characterized the intracellular folding pathway of human chorionic gonadotropin (hCG)-beta subunit in which each intermediates can be biochemically identified based on formation disulfide (S-S) bonds: p beta 1-early--> 1-late--> 2-free--> 2-combined--> native hCG-alpha beta. Based these data, we postulated that hCG-beta coincides with specific S-S bonds. We now tested this hypothesis employing Chinese hamster ovary cells transfected mutated genes Cys...

The effect of estrogen on prolactin (PRL) synthesis at a single-cell level was studied by in situ hybridization and immunohistochemistry. Long-term treatment increased PRL-containing cells from 10–20% total cell population to 80–90%, as revealed PRL mRNA containing also similar fashion. Moreover, cytoplasmic expressed the number silver grains per 4- 5-fold estrogen. These results suggest that long-term causes not only proliferation but an increase single cell.
