We provide detailed mechanisms of Ahnak-mediated potentiation transforming growth factor β (TGFβ) signaling, which leads to a negative regulation cell growth. show that Smad3 interacts with Ahnak through MH2 domain and stimulates localization into nucleus leading potentiating TGFβ-induced transcriptional activity R-Smad. Moreover, overexpression resulted in retardation cycle arrest downregulation c-Myc cyclin D1/D2. describe results from analyses Ahnak−/− mouse model expressing middle T...
NADPH oxidase (Nox) isozymes that generate intracellular reactive oxygen species (ROS) and Toll-like receptor 2 (TLR2), an inflammatory mediator, are both involved in the development of atherosclerotic lesions. To identify molecular connection between TLR2 Nox vascular remodelling, we analysed generation ROS pro-inflammatory cytokines aortic smooth muscle cells from Nox1-deficient mice response to synthetic triacylated lipoprotein Pam3CSK, a agonist. We showed signalling stimulates...
Abstract AHNAK is known to be a tumor suppressor in breast cancer due its ability activate the TGFβ signaling pathway. However, role of lung development and progression remains unknown. Here, Ahnak gene was disrupted determine effect on tumorigenesis mechanism by which it triggers investigated. First, protein expression determined decreased human adenocarcinomas compared with matched nonneoplastic tissues. Then, Ahnak−/− mice were used investigate pulmonary tumorigenesis. showed increased...
Abstract In adipose tissue, agonists of the β3-adrenergic receptor (ADRB3) regulate lipolysis, lipid oxidation, and thermogenesis. The deficiency in thermogenesis induced by neuroblast differentiation-associated protein AHNAK white tissue (WAT) mice fed a high-fat diet suggests that may stimulate energy expenditure via development beige fat. Here, we report promoted browning thermogenic gene expression WAT but not brown stimulated with ADRB3 agonist CL-316243. Consistent increased...
Ahnak protein acts as a scaffold networking phospholipase C-γ and kinase C-α, which subsequently stimulate an extracellular signal-regulated (Erk) pathway. In mouse aortic smooth muscle cells (ASMCs), the activation of signalling cascade ultimately promotes cell migration through unknown mechanism. We aimed to dissect Ahnak-mediated network involved in ASMCs.Migration ASMCs from wild-type mice was significantly increased by platelet-derived growth factor (PDGF) stimulation transwell chamber...
The AHNAK nucleoprotein has been determined to exert an anti-obesity effect in adipose tissue and further inhibit adipogenic differentiation. In this study, we examined the role of regulating hepatic lipid metabolism prevent diet-induced fatty liver. Ahnak KO mice have reportedly exhibited reduced fat accumulation liver decreased serum triglyceride (TG) levels when provided with either a normal chow diet or high-fat (HFD). Gene expression profiling was used identify novel factors that could...
Because gastrointestinal tract is not sterile, primary culture has contamination risk despite of massive washing with antimicrobial media. Microbial can play a key role in initial failure during biopsy-derived tumor culture.Tumor tissue was acquired from esophageal and gastric tumors using endoscopic biopsy. Three-dimensional cultures were performed, separated spheroids cultured media for 7 to 10 days then transferred Matrigel (Corning Inc.). We investigated factors patterns fungal...
The aim...
Development of Anthropometry-Based Equations for the Estimation Total Body Water in KoreansFor developing race-specific anthropometry-based total body water (TBW) equations, we measured TBW using bioelectrical impedance analysis (TBWBIA) 2,943 healthy Korean adults.Among them, 2,223 were used as a reference group. Two equations (TBWK1 and TBWK2) developed based on age, sex, height, weight. The adjusted R 2 was 0.908 TBWK1 0.910 TBWK2.The remaining 720 subjects validation our results.Watson...
Abstract Reactive oxygen species (ROS) play important roles as second messengers in a wide array of cellular processes including differentiation stem cells. We identified Nox4 the major ROS-generating enzyme whose expression is induced during embryoid body (EB) into cells all three germ layers. The role was examined using pluripotent (iPSCs) generated from knockout (Nox4 −/− ) mouse. Differentiation markers showed significantly reduced levels consistent with importance Nox4-generated ROS...
