A novel method, termed ladder-shape melting temperature isothermal amplification (LMTIA), was developed in this study. As a proof of concept, one pair primers or two pairs nested and thermostable DNA polymerase were employed to amplify the internal transcribed spacer Oryza sativa with curve. Our results demonstrated that LMTIA assay 50-fold more sensitive than LAMP same level specificity. The method has potential be used for prevention control emerging epidemics caused by different types pathogens.
