HSC homing, quiescence, and self-renewal depend on the bone marrow niche. A large proportion of solid tumor metastases are metastases, known to usurp homing pathways establish footholds in marrow. However, it is not clear whether tumors target niche during metastasis. Here we have shown a mouse model metastasis that human prostate cancer (PCa) cells directly compete with HSCs for occupancy Importantly, increasing size promoted metastasis, whereas decreasing compromised dissemination....

Human embryonic stem cells (hESCs) have great potentials for future cell-based therapeutics. However, their mechanosensitivity to biophysical signals from the cellular microenvironment is not well characterized. Here we introduced an effective microfabrication strategy accurate control and patterning of nanoroughness on glass surfaces. Our results demonstrated that nanotopography could provide a potent regulatory signal over different hESC behaviors, including cell morphology, adhesion,...

Abstract Nuclear receptor peroxisome proliferator-activated receptor-γ (PPARγ) is an essential transcription factor for adipocyte differentiation. In mesenchymal stem cells, PPARγ has been assumed to play a negative role in osteoblastic differentiation, by working adipogenesis dependent manner, due the reciprocal relationship between osteoblast and However, direct of function not fully understood, part inadequate model systems. Here, we describe adenoviral-mediated knockout system which...

Abstract Self-renewal of human embryonic stem cells and induced pluripotent (hiPSCs)—known as (PSC)—is influenced by culture conditions, including the substrate on which they are grown. However, details molecular mechanisms interconnecting self-renewal these remain unclear. We describe a signaling pathway in hPSCs linking expression pluripotency transcription factors to integrin α6β1 inactivation focal adhesion kinase (FAK). Disruption this results hPSC differentiation. In hPSCs, is dominant...

Bone is the preferred metastasis site of advanced prostate cancer (PCa). Using an in vivo murine model human PCa cell to bone, we noted that majority animals develop skeletal have either spinal lesions or bones hindlimb. Much less frequently, forelimb. We therefore speculated whether environment forelimb not permissive for growth PCa. Consequently, data on tumor prevalence were normalized account number cells arriving after intravascular injection, marrow cellularity, and hematopoietic stem...

There is evidence that cancer stem-like cells (CSC) and neuroendocrine behavior play critical roles in the pathogenesis clinical course of metastatic castration-resistant prostate (m-CRPC). However, there limited mechanistic understanding how CSC phenotypes impact development m-CRPC. In this study, we explored role intracellular chemokine CXCL12γ induction differentiation its on expression was detected small-cell carcinoma tissues circulating tumor from m-CRPC patients displaying an...

Growth-factor-free bone regeneration remains a challenge in craniofacial engineering. Here, we engineered an osteogenic niche composed of commercially modified alginate hydrogel and whitlockite microparticles (WHMPs), which impart tunable physicochemical properties that can direct osteogenesis human gingival mesenchymal stem cells (GMSCs). Our vitro studies demonstrate WHMPs induce GMSCs more effectively than previously demonstrated hydroxyapatite (HApMPs). Alginate–WHMP hydrogels showed...

Prostate cancer metastases and hematopoietic stem cells (HSC) frequently home to the bone marrow, where they compete occupy same HSC niche. We have also shown that under conditions of stress, HSCs secrete morphogenetic proteins (BMP)-2 BMP-6 drives osteoblastic differentiation from mesenchymal precursors. As it is not known, we examined whether metastatic prostate can alter regulation normal formation by progenitor (HPC). HSC/HPCs isolated mice bearing nonmetastatic tumor were their ability...

Regulation of mTOR signaling depends on an intricate interplay post-translational protein modification. Recently, mEAK-7 (mTOR associated protein, eak-7 homolog) was identified as a positive activator via alternative complex. However, the upstream regulation in human cells is not known. Because microRNAs are capable modulating translation RNA eukaryotes, we conducted bioinformatic search for relevant targeting using Exiqon miRSearch V3.0 algorithm.
