A general and sensitive nonempirical protocol to determine the absolute configurations of erythro threo diols, amino alcohols, diamines is reported. Binding diols porphyrin tweezer system greatly enhanced by increasing Lewis acidity metalloporphyrin. Supramolecular complexes formed between host chiral substrates exhibited exciton-coupled bisignate CD spectra with predictable signs based on substituents center. The working model suggests that observed helicity dictated via steric...
A novel fluorescent cassette consisting of a TPE donor and cyanine acceptor was developed for sensitive detection pH changes in live cells.
Abstract Determining the absolute stereochemisty of small molecules bearing remote nonfunctionalizable stereocenters is a challenging task. Presented solution in which appropriately substituted bis(porphyrin) tweezers are used. Complexation suitably derivatized β‐, γ‐, or δ‐chiral carboxylic acid to tweezer induces predictable helicity bis(porphyrin), detected as bisignate Cotton Effect (ECCD). The sign ECCD curve correlated with stereochemistry substrate based on derived working mnemonics manner.
The synthesis of oligodeoxynucleotides (ODNs) containing the sensitive N6-acetyladenosine (6acA), N2-acetylguanosine (2acG), and N4-methyoxycarbonyldeoxycytidine (4mcC), as well up to four N4-acetyldeoxycytidine (4acC) modifications is described. meDmoc group was used for protection exo-amino groups nucleobases, Dmoc linker solid phase synthesis. Deprotection cleavage were achieved under non-nucleophilic conditions, which highly 4acC, 6acA, 2acG, 4mcC found completely stable. Among modified...
Abstract Metallated porphyrin tweezers have demonstrated a remarkable ability to function as reporters of absolute stereochemistry for number different classes organic molecules. Flexibility in binding, however, can result an ensemble Exciton Coupled Circular Dichroism (ECCD) active conformations that could lead variable results. Linker flexibility was found be key determinant binding conformation. Experimental results indicate balance between linker and rigidity yield optimum tweezer...
Chemical cross-linking combined with proteolytic digestion and mass spectrometry (MS) is a promising approach to provide inter- intramolecular distance constraints for the structural characterization of protein topologies functional multiprotein complexes. Despite relative straightforwardness these methodologies, identification cross-linked proteins presents significant analytical challenge, due complexity resultant peptide mixtures, as well array inter-, intra-, or "dead-end"-cross-linked...
We have developed two highly sensitive cyanine dyes, which we refer to as probes
The specificity of carbohydrate transporters towards their substrates poses a significant challenge for the development molecular probes to monitor sugar uptake in cells biochemical and biomedical applications. Herein we report new set coumarin-based fluorescent conjugates applicable analysis fructose due free passage through fructose-specific transporter GLUT5. reported cover broad range fluorescence spectrum providing essential tools evaluation transport capacity live cells.
Two near-infrared fluorescent probes (A and B) containing hemicyanine structures appended to dipicolylamine (DPA), a derivative where one pyridine was substituted with pyrazine, respectively, were synthesized tested for the identification of Zn(II) ions in live cells. In both probes, an acetyl group is attached phenolic oxygen atom platform decrease probe fluorescence background. Probe A displays sensitive responses binds preferentially over other metal such as Cd2+ low detection limit 0.45...
Abstract Recent publications suggest that high dietary fructose might play a significant role in cancer metabolism and can exacerbate number of aspects metabolic syndrome. Addressing the plays human health is controversial question requires detailed understanding many factors including mechanism transport into healthy diseased cells. Fructose cells thought to be largely mediated by passive hexose transporters Glut2 Glut5. To date, no probes selectively transported one these enzymes but not...
Point-of-care applications rely on biomedical sensors to enable rapid detection with high sensitivity and selectivity. Despite advances in sensor development, there are challenges cancer diagnostics. Detection of biomarkers, cell receptors, circulating tumor cells, gene identification, fluorescent tagging time-consuming due the sample preparation response time involved. Here, we present a novel approach target enhanced metabolism breast cancers for using imaging. Fluorescent analogs fructose...
The catching-by-polymerization (CBP) oligodeoxynucleotide (oligo or ODN) purification method has been demonstrated suitable for large-scale, parallel, and long oligo purification. authenticity of the oligos verified via DNA sequencing. Gene construction expression have demonstrated. A remaining obstacle to practical utility CBP is affordable polymerizable tagging phosphoramidites (PTPs) that are needed method. In this article, we report scalable synthesis four nucleoside (dA, dC, dG dT)...
Abstract Porphyrin tweezers have been successfully used as hosts for the absolute stereochemical determination of a variety chiral compounds. A set new porphyrin with substituted aryl groups on meso position rings synthesized. The modified are diamines and carrier‐derivatized α‐chiral carboxylic acids in order to monitor influence various substitutions group amplitude sign ECCD couplet. t ‐Butyl substitution at meta positions porphyrin's phenyl substituents leads enhanced amplitudes....
Three fluorescent probes A, B, and C that function in the near-infrared wavelengths utilize pseudo xanthene platforms with an oxygen atom at 10-position replaced by a [Me-N]2– group have been created to identify hypoxia via nitroreductase determinations 0.04, 0.10, 0.19 ng/mL levels. Theoretical calculations suggest are not planar due steric interactions. Absorptions of photons result transition electron density from indoline moieties delocalized orbitals on anthranilic section, ending up...
Abstract CD and NMR spectroscopic evidence of cooperative binding between an α‐halogen atom a carboxamide group with zinc porphyrin leads to unprecedented conformation for the determination absolute stereochemistry α‐haloamides (α‐halocarboxylic acids derivatized 1,4‐phenylenediamine) through use exciton‐coupled circular dichroism (ECCD). With chiral lactams, whose rotomeric contributions are minimized, both ECCD spectroscopy demonstrate that favors side sterically more demanding halogen as...
Abstract Coumarins are widely applied in sensing devices due to their diverse fluorescence properties. In this paper, we consider the C4‐substituted 7‐aminocoumarins investigate structure‐property relationship via examining effect of various substitutions on optical Absorption and measurements together with time‐dependent density functional theory calculations described. We find that significant red shift spectra is associated cyclic carbonyl group coumarins a substantial increase dipole...
