In cross-sectional studies, elevated plasma homocysteine levels have been associated with poor cognition and dementia. Studies of newly diagnosed dementia are required in order to establish whether the precede onset or result from dementia-related nutritional vitamin deficiencies.

Background Methylenetetrahydrofolate reductase (MTHFR) synthesizes 5-methyltetrahydrofolate, the major carbon donor in remethylation of homocysteine to methionine. A common MTHFR mutation, an alanine-to-valine substitution, renders enzyme thermolabile and may cause elevated plasma levels amino acid homocysteine. Methods Results To assess potential interaction between this mutation vitamin coenzymes metabolism, we screened 365 individuals from NHLBI Family Heart Study. Among with lower folate...

Epidemiologic studies have identified hyperhomocysteinemia as a possible risk factor for atherosclerosis. We determined the of carotid-artery atherosclerosis in relation to both plasma homocysteine concentrations and nutritional determinants hyperhomocysteinemia.

DNA methylation, an essential epigenetic feature of that modulates gene expression and genomic integrity, is catalyzed by methyltransferases use the universal methyl donor S -adenosyl- l -methionine. Methylenetetrahydrofolate reductase (MTHFR) catalyzes synthesis 5-methyltetrahydrofolate (5-methylTHF), for methionine from homocysteine precursor In present study we sought to determine effect folate status on methylation with emphasis interaction common C677T mutation in MTHFR gene. A liquid...

The cytokines IL-1 beta and TNF-alpha cause cachexia hypermetabolism in animal models, but their role human inflammation remains controversial. relationship between vitro cytokine production metabolism was examined 23 adults with RA healthy control subjects matched on age, sex, race, weight. Body composition measured by multicompartmental analysis of body cell mass, water, fat, bone mass. Resting energy expenditure (REE) indirect calorimetry. Cytokine PBMC radioimmunoassay. Usual intake,...

Abstract Nationwide fortification of enriched uncooked cereal grains with folic acid began in the United States and Canada 1996 1997, respectively, became mandatory 1998. The rationale was to reduce number births complicated by neural tube defects. Concurrently, experienced abrupt reversals downward trend colorectal cancer (CRC) incidence that two countries had enjoyed preceding decade: absolute rates CRC increase (United States) 1998 (Canada), peaked 2000 have continued exceed pre-1996/1997...

Background: Total homocysteine levels are associated with arteriosclerotic outcomes. Objective: To determine whether total predict incident stroke in elderly persons. Design: Prospective population-based cohort study 9.9 years of follow-up. Setting: Framingham, Massachusetts. Patients: 1947 Framingham Study participants (1158 women and 789 men; mean age ± SD, 70 7 years). Measurements: Baseline 9.9-year incidence. Results: The quartiles nonfasting were as follows: quartile 1, 4.13 to 9.25...

Background: The concentration of circulating total homocysteine is a sensitive marker inadequate folate and vitamin B12 status. Elevated concentrations are associated with an increased risk for vascular disease. Objective: To identify reference ranges serum in U.S. residents quantify the contribution to high concentrations. Design: Cross-sectional prevalence study. Setting: United States. Patients: A nationally representative sample 3563 male participants 4523 female 12 years age or older...

Abstract There is an excess prevalence of hyperhomocysteinemia in dialysis-dependent end-stage renal disease (ESRD) patients. Cross-sectional studies the relationship between elevated total homocysteine (tHcy) levels and prevalent cardiovascular (CVD) this patient population suffer from severe methodologic limitations. No prospective investigations examining association tHcy subsequent development arteriosclerotic CVD outcomes among maintenance dialysis patients have been reported. To assess...
