In this study, we show that oligodendrocyte differentiation is powerfully inhibited by activation of the Notch pathway. Oligodendrocytes and their precursors in developing rat optic nerve express Notch1 receptors and, at same time, retinal ganglion cells Jagged1, a ligand receptor, along axons. Jagged1 expression developmentally regulated, decreasing with time course parallels myelination nerve. These results suggest timing controlled pathway raise question whether localization

Recombinant adeno-associated virus (rAAV) vectors are a leading gene delivery platform, but vector manufacturing remains challenge. New methods needed to increase rAAV yields and reduce costs. Past efforts improve production have focused on optimizing single variable at time, this approach does not account for the interactions of multiple factors that contribute generation. Here, we utilized design-of-experiment (DOE) methodology optimize in HEK293T suspension cell system. We simultaneously...

Abstract Background Elucidation of immune populations with single-cell RNA-seq has greatly benefited the field immunology by deepening characterization heterogeneity and leading to discovery new subtypes. However, methods inherently suffer from limitations in recovery complete transcriptomes due prevalence cellular transcriptional dropout events. This issue is often compounded limited sample availability prior knowledge heterogeneity, which can confound data interpretation. Results Here, we...

The axial orientation of molecular chains always results in an increase fiber strength and a decrease toughness. Here, taking inspiration from the skin structure, artificial spider silk with buckled sheath-core structure is developed, mechanical toughness reaching 1.61 GPa 466 MJ m-3 , respectively, exceeding those Caerostris darwini silk. achieved by nano-pulley combing polyrotaxane hydrogel fibers through cyclic stretch-release training, which exhibits alignment polymer core buckling...

As the most aggressive brain tumor, chemotherapy of malignant glioma remains to be extremely challenging in clinic. The blood-brain barrier (BBB) and tumor (BBTB) are physiological pathological barriers preventing therapeutic drugs from reaching region. In addition, vasculogenic mimicry (VM) formed by invasive cells instead endothelial angiogenesis very common glioma, leading poor prognosis recurrence glioma. An ideal drug delivery system for needs traverse BBB BBTB then target VM,...

We report the implementation of a parallel microscopy system (96 Eyes) that is capable simultaneous imaging all wells on 96-well plate. The optical consists 96 units, where each unit made out four element objective, through molded injection process, and low cost CMOS camera chip. By illuminating sample with angle varying light applying Fourier Ptychography, we can improve effective brightfield numerical aperture objectives from 0.23 to 0.3, extend depth field ±5 μm ±15 μm. use Ptychography...

Autophagy is the primary process for recycling cellular constituents through lysosomal degradation. In addition to nonselective autophagic engulfment of cytoplasm, autophagosomes can recognize specific cargo by interacting with ubiquitin-binding autophagy receptors such as SQSTM1/p62 (sequestosome 1). This selective form important degrading aggregation-prone proteins prominent in many neurodegenerative diseases. We carried out a high content image-based siRNA screen (4 8 per gene) modulators...

Tumor-homing peptides have been widely used to mediate active targeted drug delivery. l-AE is a reported targeting peptide demonstrating high binding affinity epidermal growth factor receptor (EGFR) and mutation variant III (EGFRvIII) overexpressed on neovasculature, vasculogenic mimicry, tumor cells, stem cells. To improve its proteolytic stability, d-peptide ligand (termed d-AE, the enantiomer of l-AE) was developed. d-AE confirmed bind receptors EGFR EGFRvIII with capability comparable...

Adeno-associated virus (AAV) has been used extensively as a vector for gene therapy. Despite its widespread use, the mechanisms by which AAV enters cell and is trafficked to nucleus are poorly understood. In this study, we performed two pooled, genome-wide screens identify positive negative factors modulating AAV2 transduction. Genome-wide libraries directed against all human genes with four designs per or eight were transduced into U-2 OS cells. These pools encoding EGFP sorted based on...

Folic acid (FA) is one of the most widely utilized small-molecule ligands for cancer targeted drug delivery. Natural IgM was recently found to avidly absorb on surface FA-functionalized liposomes (FA-sLip), negatively regulating in vivo performance by efficiently activating complement. Herein, lipodiscs (FA-Disc) were constructed successfully circumvent IgM-mediated opsonization and retained binding activity with folate receptors vivo. The FA moiety along bound restricted highly curved rim...
